人乳腺组织性激素代谢相关酶表达的相关研究
Correlational research on expressions of sexual hormone metabolism related enzymes in human breast tissue
【摘要】 目的:探讨参与雌酮(Estrone,E1)和雌二醇(Estradiol,E2)相互转变的酶即17βHSD12、17βHSD7、17βHSD1、17βHSD2在乳腺良、恶性肿瘤组织内的表达及其与病理特点的关系。方法:采用RT-PCR方法测定17βHSD12、17βHSD7、17βHSD1、17βHSD2在48例女性乳腺良性及恶性肿瘤组织中的表达情况,并分析该类酶与临床病理参数的关系。结果:17βHSD12在乳腺癌组织高表达(96.96%),绝经后乳腺癌组织中的表达率为100.00%,与良性肿瘤组织表达率66.67%相比差异有统计学意义(P<0.05);17βHSD12表达率与癌组织雌激素受体(ER)、孕激素受体(PR)、人表皮生长因子受体-2(Her-2)、淋巴结转移与否无关;17βHSD1、17βHSD7、17βHSD2在良、恶性肿瘤组织表达差异无统计学意义(P>0.05)。结论:17βHSD12与乳腺组织的恶性转化有关,有望成为乳腺癌内分泌治疗新靶点。
【Abstract】 Objective: To explore the expressions of enzymes engaged in conversion between estrone and estradiol( 17βHSD12, 17βHSD7,17βHSD1,17βHSD2) in benign or malignant breast tissues and their relationships with clinicopathological characteristics. Methods: The expression rates of 17βHSD12,17βHSD7,17βHSD1,17βHSD2 in 48 female benign and malignant breast tumor specimens were detected by RT- PCR. The relationship between the expressions and clinicopathological parameters was analyzed. Results: The expression rates of 17βHSD12 in breast cancer tissue and postmenopausal breast cancer tissue were 96. 96% and 100. 00%,respectively,compared with benign breast tumor tissue( 66. 67%),there was statistically significant difference( P < 0. 05); the expression rate of 17βHSD12 was not correlated with estrogen receptor,progesterone receptor,Her- 2,and lymph node metastasis; there was no statistically significant difference in the expression rates of 17βHSD1,17βHSD7,and 17βHSD2 between benign and malignant tumor tissues( P > 0. 05). Conclusion: 17βHSD12 is related to malignant transformation of breast tissue,which may be a new target of endocrine therapy.
