FOXP1在人卵巢癌细胞系C13~*和OV2008中的表达及其意义
Expression and significance of FOXP1 in human ovarian cancer cell lines C13* and OV2008
【摘要】 目的:研究FOXP1在人卵巢癌顺铂耐药细胞株C13*和敏感细胞株OV2008的表达和细胞定位情况,并初步探讨其表达意义。方法:①流式细胞术检测不同浓度顺铂处理后两株细胞的凋亡率;②实时荧光定量PCR和免疫荧光技术分别从基因和蛋白水平检测FOXP1在两株细胞中的表达和细胞定位情况。结果:①C13*对顺铂敏感性低于OV2008(P<0.05);②实时荧光定量PCR显示:FOXP1在C13*中的表达明显低于在OV2008中的表达(P<0.05);③免疫荧光显示:FOXP1在两株细胞胞核中均100%表达,而在C13*胞浆中的表达显著低于在OV2008胞浆中的表达(P<0.05),同时大部分OV2008胞浆呈现强"帽子"样染色,而此染色现象在C13*中非常微弱甚至消失。结论:FOXP1在卵巢癌细胞胞浆和胞核中均可表达,而在C13*胞浆的表达下调可能与其耐药有关。
【Abstract】 Objective: To study the expression and cellular localization of FOXP1 in cisplatin-resistant ovarian cancer cell line C13*and cisplatin-sensitive ovarian cancer cell line OV2008,and to investigate its significance. Methods: ① Cultured C13*and OV2008 were treated with different concentrations of cisplatin followed by detection of cell apoptosis rate using flow cytometry. ②Real-time fluorescence quantitative PCR and immunofluorescence technique were used to detect the gene and protein expression level of FOXP1 in the two cell lines,respectively. Results: ①Cisplatin sensitivity of C13*was lower than that of OV2008( P < 0. 05). ②The results of real-time fluorescence quantitative PCR showed that the gene expression level of FOXP1 in C13*was lower than that in OV2008( P < 0. 05). ③The results of immunofluorescence implied that the expression of FOXP1 was found in all nucleus of both C13*and OV2008 cells( P >0. 05),but the expression of FOXP1 in cytoplasm of C13*was lower and weaker than that of OV2008( P < 0. 05),and the staining of cytoplasm in OV2008 looked like the structure of " cap",but this phenomenon in C13*was very weak. Conclusion: The expression of FOXP1 can be found both in nucleus and cytoplasm of ovarian cancer cell and the down-regulation of FOXP1 in cytoplasm of C13*may be related with drug resistance.
