应用DGC结合MACS技术优选精子的效果评价
Evaluation of combination of MACS with DGC on sperm optimization
【摘要】 目的:评价密度梯度离心(DGC)结合磁性活性细胞分选法(MACS)优选精子质量的效果。方法:应用DGC结合MACS技术优选精液标本40份,比较优选前、DGC优选及DGC结合MACS技术优选后精子活率、前向精子活动力、正常形态百分率、DNA损伤的变化。结果:DGC结合MACS技术优选后,精子活率、前向活动力及DNA完整性较优选前原液及单用DGC法优选均明显提高(P<0.01)。结论:DGC结合MACS技术可以优选出精子活率、活动力、形态较好而DNA结构受损比例较小的精子群体,可作为一种优良的精子优选方法。
【Abstract】 Objective: To evaluate the combination of magnetic activated cell sorting( MACS) and density gradient centrifugation( DGC) in optimization of semen quality. Methods: Forty semen samples were prepared by MACS following DGC. The changes of sperm viability,motility,morphology and sperm DNA integrity were compared to evaluate the combined technique in optimization of semen quality. Results: Spermatozoa prepared by MACS following DGC had higher viability,motility and DNA integrity values when compared with either spermatozoa prepared by DGC only or raw semen samples( P < 0. 01). Conclusion: As a good method of sperm selection,the combination of DGC and MACS could optimize superior spermatozoa in terms of sperm viability,motility,morphology and DNA integrity.
