【摘要】 目的观察促红细胞生成素(EPO)对慢性马兜铃酸肾病(CAAN)大鼠血管性假血友病因子(vWF)表达的影响,探讨促红细胞生成素对慢性马兜铃酸肾病大鼠的肾脏保护作用。方法雌性Wistar大鼠30只,随机分三组:①CAAN模型组,②EPO治疗组,③正常对照组,每组各10只,关木通水煎剂灌胃12周制备CAAN大鼠模型。从第13周起,治疗组大鼠给予皮下注射重组人EPO每次50 IU/kg,每周3次,共6周。第19周检测各组大鼠血尿素氮(BUN)、血肌酐(SCr)、尿蛋白、血红蛋白(Hb)、血vWF水平;免疫组化法检测肾组织vWF的表达。结果与CAAN组比较,EPO治疗组BUN、SCr、尿蛋白水平、vWF水平下降,Hb水平升高(均P<0.05)。同时EPO治疗组大鼠肾组织vWF的表达减少(P<0.05)。结论 EPO治疗可以改善CAAN大鼠的肾功能,可能与其下调vWF,减轻内皮损伤有关。更多还原

【Abstract】 Objective To observe the effect of erythropoietin on the expression of Von Willebrand factor(vWF) in CAAN rats and to investigate the protective effect of erythropoietin of kidney in rats with CAAN. Methods Thirty female Wistar rats were randomLy divided into 3 groups. ①CAAN group(n=10); ②EPO group(n=10); ③SHAM group(n=10); CAAN group and EPO group were infused into stomachs with Caulis aristolochia manshuriensis decoction for 12 weeks. From week 13,EPO group were received EPO by hypodermic injection with 50 IU/kg three times a week for 6 weeks. At weeks 19,All the rats were killed. Specimens of blood and urine were collected to detect the blood urea nitrogen(BUN),Serum creatinie(Scr), Haemato-globin(Hb),urine protein and levels of vWF. HE and Masson staining and micro-scopy were used to observe the pathology of the kidney. Immunohistochemistry were used to detect the expression of vWF. Results Urinary protein, serum creatinine and BUN and levels of vWF in EPO treated group were much lower than those of CAAN group; Hb was increased(both P<0.05). The A value of vWF+decreased in EPO treated group comparing to CAAN group(P<0.01). Conclusion EPO could deduce the level of vWF and decreased thrombosis.更多还原
