孔雀石绿及其代谢物在杂交鳢(斑鳢♀×乌鳢♂)体内的残留及消除规律研究

【摘要】 本实验模拟自然养殖条件,对杂交鳢(斑鳢Channa maculata♀×乌鳢Channa argus♂)肌肉中的孔雀石绿(MG)及其代谢物隐色孔雀石绿(LMG)的残留及消除规律进行了研究。对池塘中初始体重为(200±10)g的杂交鳢进行质量浓度为200μg/L的MG药浴,药浴后于0.5、1、2、4、8、12、24、48、96、192、288、384、480、720、960、1 200、1 440、1 920、2 400、2 880、3 600、4 320、5 040和5 760 h取样,采用液相色谱串联质谱法(HPLC-MS/MS)测定杂交鳢肌肉组织中MG和LMG的残留量。药浴0.596 h期间,MG和LMG在杂交鳢肌肉中的残留量不断波动式升高,分别达到(543.77±22.14)μg/kg和(220.51±7.93)μg/kg。MG从96 h之后呈现下降趋势,到1 920 h后MG残留量低于检测限0.5μg/kg。而LMG在288480 h范围内的残留量基本维持在(73.72±4.85)70.33±9.27μg/kg之间,1 920h时LMG的残留量急剧下降至(2.25±0.25)μg/kg,至5 040 h已低于检测限0.5μg/kg。结果表明,MG和LMG在杂交鳢体内的残留消除期远大于水体,且LMG比MG代谢时间长。

【Abstract】 The residues and depuration of malachite green( MG) and its primary metabolite leuco- malachite green( LMG) in hybrid snakeheads( Channa maculata ♀ × Channa argus ♂) were studied. The average weight of hybrid snakeheads was about( 200 ± 10) g,and they were exposed to 200 μg / L MG in a 667 m2 pond. The samples were collected at 0. 5,1,2,4,8,12,24,48,96,192,288,384,480,720,960,1 200,1 440,1 920,2 400,2 880, 3 600,4 320,5 040,and 5 760 h respectively,and the residues of MG and LMG were tested by high performance liquid chromatography tandem mass spectrometry( HPLC- MS / MS). The residues of MG and LMG increased in muscle unceasingly from 0. 5 to 96 h and were up to( 543. 77 ± 22. 14) μg / kg and( 220. 51 ± 7. 93) μg / kg,respectively. The residues of MG decreased after 96 h,and were below the detection limit of 0. 5 μg / kg at 1 920 h. The residue of LMG maintained between( 73. 72 ± 4. 85) and( 70. 33 ± 9. 27) μg / kg from 288 to 480 h,while significantly decreased to( 2. 25 ± 0. 25) μg / kg at 1 920 h,and was below the detection limit of 0. 5 μg / kg at 5 040 h. The results also indicated that LMG was more slowly degradated than MG,which could be used as a marker to determine whether MG was used in aquaculture.
