β-乳球蛋白共轭亚油酸复合物对LoVo细胞凋亡的影响

【摘要】 目的研究β-乳球蛋白共轭亚油酸复合物(β-lactoglobulin conjugated linoleic acid complex,β-LG-CLA)对人结肠癌LoVo细胞的凋亡诱导作用。方法 WST-1法分析复合物对LoVo细胞增殖的影响;倒置显微镜观察复合物对细胞形态学改变;JC-1染色检测细胞线粒体膜电位变化;caspase-3蛋白活性测定。结果β-LG-CLA可抑制LoVo细胞增殖,抑制率呈时间剂量依赖性;β-LG-CLA作用于结肠癌LoVo细胞,使结肠癌细胞出现了细胞数量明显减少,体积缩小,皱缩变形,进一步有凋亡小体产生,随着β-LG-CLA浓度的增加,此现象更加明显;β-LG-CLA剂量组细胞线粒体膜电位降低,呈现浓度依赖性;β-LG-CLA剂量组细胞caspase-3蛋白活性含量比对照组高,且呈现剂量-时间反应关系。结论β-LG-CLA可诱导结肠癌LoVo细胞凋亡,表现为细胞内线粒体膜电位降低以及释放性蛋白caspase-3活性升高。

【Abstract】 Objective To investigate the effects of β-lactoglobulin and conjugated linoleic acid complex(β-LGCLA) on apoptosis in LoVo colon cancer cells. Methods WST-1 assay was used to examine the effects of β-LG-CLA on the proliferation of LoVo cells. The morphology of LoVo cells was observed under inverted microscope. The mitochondrial membrane potential was detected by the JC-1staining. Caspase-3 protein activity was also measured. Results β-LG-CLA inhibited the proliferation of LoVo cells dose- and time-dependently. Typical apoptotic cells were observed. The number of viable cells was decreased, and the volume shrunk and apoptotic bodies appeared. These changes were more significant with increaseing concentration of β-LG-CLA. The mitochondrial membrane potential was decreased in a dose-dependent manner. Caspase-3 protein activity was increased dose- and time-depenclently. Conclusion β-LG-CLA induces the apoptosis of LoVo cells, as indicated by the reduction of mitochondrial membrane potential and elevation of caspase-3 activity.
