大孔树脂分离黄杞叶总黄酮的研究

【摘要】 目的通过研究确定黄杞叶总黄酮大孔树脂吸附工艺参数。方法通过大孔吸附树脂对黄杞叶黄酮成分的吸附-洗脱性考察,来确定各工艺参数。结果 D101树脂对黄杞叶总黄酮具有较好分离作用,黄杞叶总黄酮动态饱和吸附量为55 mg/g;当药材树脂比为1∶2,树脂柱径高比为1∶7,上样药液为水提液,浓度相当于生药浓度0.4 g/m L,以水、10%乙醇、60%乙醇进行洗脱,洗脱量分别为10、10和5倍树脂柱量,收集60%醇洗部分,减压浓缩至干,即得黄杞叶总黄酮提取物。结论采用D101树脂对黄杞叶总黄酮具有较好分离作用,总黄酮回收率在90%以上。

【Abstract】 Objective To get an efficient preparative separation process of flavonoid crude extracts of leaves of Engelhardtia roxburghiana wall. Method The optimum resin was to chosen through the researches of their character of adsorption and desorption of FEE. Results D101 resin offers the best adsorption capacity, and adsorption and desorption ratios. The dynamic adsorption capacity D101 resin was 55 mg/g. The adsorption and desorption experiments have been carried out on a D101 resin packed column to get the optimal separation process of FEE. The crude material and resin ratio is 1∶2, the hight and diameter ratio is 1∶7, the solution concentration equal to crude material is 0.4 g/m L. The aquatic extract was applied to column, washed by sixcolumn volumes of water, 10 sixcolumn volumes of 10% alcohol, 5 sixcolumn volumes of 60% alcohol successively. The part of 60% alcohol was collected and concentrated by vacuum drying method to get FEE. Conclusions D101 resin offers the best adsorption capacity, and adsorption and desorption ratios of FEE. This D101 resin method provides a potential approach for large-scale separation and purification of FEE with recovery of 90% higher.
