组织蛋白酶L mRNA在大鼠早期心肌缺血组织中的表达及其意义

【摘要】 目的探讨组织蛋白酶L(cathepsin L)基因在早期心肌缺血组织中的表达及其意义。方法运用结扎冠状动脉左前降支的方法制作心肌缺血动物模型。健康SD大鼠随机分为手术组、假手术组和非手术对照组,并将手术组心肌标本按缺血区和非缺血区又分为心肌缺血组和非缺血组,手术组和假手术组大鼠按冠状动脉结扎时间段不同又分为0、15、30、60和120 min五个亚组,通过时聚合酶链反应(real time polymerase chain reaction,RTPCR)测定每个样本组织蛋白酶L的表达水平,同时以大鼠β-actin基因作为内参对照。结果①心肌缺血组组织蛋白酶L在120 min的表达明显低于30和60 min两个时间点,差异有统计学意义(P<0.05);②非缺血组大鼠手术后15 min组织蛋白酶L的表达明显高于假手术组,差异有统计学意义(P<0.05)。结论 cathepsin L基因可能参与早期缺血心肌的病理生理调节过程,对该基因的进一步研究,有望在对心肌早期缺血发病机制的认识、心肌缺血的诊断和治疗以及为心源性猝死的法医学诊断方面提供理论依据。

【Abstract】 Objective To investigate the Cathepsin L gene expression in early ischemic myocardium in rats. Methods Adult healthy Sprague-Dawley( SD) rats were randomly divided into four groups: ischemia,non-ischemia,sham-operated and normal control groups. The expressions of Cathepsin L and β-actin genes in myocardium was examined by a real time PCR at 0 min,15 min,30 min,60 min and 120 min after ligation of left anterior descending coronary artery. Results It was found that ①the expression of Cathepsin L gene at the 120 min after ischemia was lesser than that in the 30 min and 60 min( P < 0. 05) and ②15 min after the injury,the expression of Cathepsin L gene in the non-ischemia group was significantly higher than that in the sham-operated group. Conclusion Our findings indicate that Cathepsin L gene may participate in pathophysiologic regulating process at the early stage of myocardial ischemia in rats.
