枸杞多糖对2型糖尿病大鼠视网膜炎症因子表达的影响

【摘要】 目的探讨枸杞多糖对2型糖尿病大鼠模型视网膜炎症因子表达的影响。方法大鼠随机分为正常对照组,模型对照组,盐酸二甲双胍阳性对照组,枸杞多糖(LBP)高、中、低剂量组。除正常对照组外,其余各组均以高糖高脂饲料加链脲佐菌素复制2型糖尿病大鼠模型,治疗结束后,用酶联免疫(ELISA)法检测各组大鼠视网膜组织中肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6),C反应蛋白(CRP)浓度。结果与正常对照组相比,2型糖尿病模型组大鼠视网膜TNF-α,IL-6,CRP浓度均明显升高(P<0.05);与2型糖尿病模型组比较,LBP高剂量组大鼠视网膜TNF-α浓度显著降低(P<0.01),LBP高、中剂量组大鼠视网膜IL-6浓度明显降低(P<0.05),LBP高、中剂量组大鼠视网膜CRP浓度显著降低(P<0.01)。结论 LBP可能通过调节视网膜组织的TNF-α,IL-6,CRP水平,减少血-视网膜屏障炎症因子的表达,从而有效改善2型糖尿病视网膜病变炎症。

【Abstract】 Objective To investigate the influence of lycium barbarum polysaccharides(LBP) on the expression of the inflammatory cytokines in the retina of type 2 diabetes mellitus(T2DM) rats model.Methods The rats were randomly divided into the normal control group,model control group,metformin hydrochloride positive control group,high-,medium-and low-dose of LBP groups.Except the normal control group,the other groups were fed with high-sugar,high-lipid fodder combined with streptozotocin to copy T2DM rat models.After the medication,ELISA was used to detect the expression of TNF-α,IL-6 and CRP in rat retina.Results Compared with the normal control group,the retinal levels of TNF-α,IL-6 and CRP were significantly increased in the T2DM model group(P < 0.05).Compared with the T2DM model group,the retinal level of TNF-α in the high-dose LBP group was significantly decreased(P < 0.01),the retinal level of IL-6 in the high-dose and medium-dose LBP groups was significantly decreased(P < 0.05),the retinal level of CRP in the high-dose and medium-dose LBP groups was significantly decreased(P < 0.01).Conclusion LBP may effectively improve the inflammation symptoms of T2DM retinopathy by adjusting the retinal levels of TNF-α,IL-6 and CRP and decreasing the expression of the inflammatory cytokines.
