波长切换高效液相色谱法同时测定化胃舒颗粒中4种有效成分的含量

【摘要】 目的建立同时测定化胃舒颗粒中毛蕊异黄酮苷、阿魏酸、甘草苷和橙皮苷4种成分含量的反相高效液相色谱法。方法采用Synergi C18柱(250 mm×4.6 mm,4μm),以乙腈-0.05%磷酸水溶液(体积分数)为流动相进行梯度洗脱,流速为1.0 mL/min,柱温为30℃。采用切换波长的方法,在260 nm下检测毛蕊异黄酮苷,在316 nm下检测阿魏酸,在237 nm下检测甘草苷,在280 nm下检测橙皮苷。结果毛蕊苷异黄酮苷、甘草苷、阿魏酸和橙皮苷的进样量分别在0.087 5~0.437 8μg(r=1,n=5),0.085 45~0.427 3μg(r=0.999 8,n=5),0.081 6~0.408μg(r=0.999 9,n=5)和0.123 5~0.618μg(r=0.999 8,n=5)范围内与峰面积呈良好的线性关系,平均加样回收率依次为96.84%,98.04%,97.35%和98.10%,RSD分别为0.98%,1.28%,1.31%和1.05%。结论该方法可同时测定化胃舒颗粒中毛蕊苷异黄酮苷、甘草苷、阿魏酸和橙皮苷的含量。

【Abstract】 Objective To develop a RP-HPLC method for the content determination of calycosin-7-glucoside,liquiritin,ferulic acid and hesperidin in Huaweishu Granules. Methods HPLC was performed on the Synergi C18 column( 250mm × 4. 6 mm,4μm) with the gradient elution of acetonitrile-0. 05% phosphoric acid aqueous solution at a flow rate of 1. 0 mL/min. The column temperature was maintained at 30 ℃. The wavelength switching method was adopted for conducting the detection at 260 nm for calycosin-7-glucoside,at 237 nm for liquiritin,at 316 nm for ferulic acid and at 280 nm for hesperidin. Results The linear ranges were 0. 087 5-0. 437 8 μg( r = 1,n = 5),0. 085 45-0. 427 3 μg( r = 0. 999 8,n = 5),0. 0816-0. 408 μg( r = 0. 999 9,n = 5) and 0. 123 5-0. 618 μg( r = 0. 999 8,n = 5),respectively. The recovery rates were 96. 84%( RSD = 0. 98%),98. 04%( RSD = 1. 28%),97. 35%( RSD = 1. 31%) and 98. 10%( RSD = 1. 05%),respectively. Conclusion This method can simultaneously determine the contents of calycosin-7-glucoside,liquiritin,ferulic acid and hesperidin in Huaweishu Granule.
