两种抗菌肽的体内外抗菌功能研究
In vitro and In vivo Antimicrobial Functions of Two Antimicrobial Peptides
【摘要】 通过对抗菌肽的一系列修饰,包括缩短长度、增加抗菌能力以及延长其体内半衰期来降低抗菌肽的生产成本。基于抗菌肽LL-37的氨基酸序列,对抗菌肽进行结构与功能改造,对改造抗菌肽的抗菌谱、抗菌能力、体外血浆中的抗菌动力学以及其体内抗菌功能进行了详细研究。成功改造了2种抗菌肽,分别由21和29个氨基酸组成,它们具有强的体内外抗菌活性和广的抗菌谱。改造的抗菌肽抗菌能力更强,生产成本更低。
【Abstract】 The aim of our present research is to decrease the production expense of antimicrobial peptides. The length of antimicrobial peptides was planosed to shorten and increase their antimicrobial potential and prolong their half life in vivo. Based on the sequence of human cathelicidin antimicrobial peptide,LL-37,amino acid residues were replaced and sequence length was shortened. The in vitro and in vivo antimicrobial abilities of these mutants were tested. The results showed that the LL-21 and LL-29 two mutants with improved antimicrobial abilities and shorter amino sequences were obtained. These two modified LL-37 mutants had stronger antimicrobial abilities and cheaper production expenses than that of their native counterparts.
