毛两面针药材HPLC指纹图谱及模式识别研究

【摘要】 目的:通过HPLC法建立毛两面针药材指纹图谱评价体系,为控制其质量提供有效、可靠的实验依据。方法:采用Promosil C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈(A)-0.2%磷酸加0.2%三乙胺水溶液(B)为流动相,梯度洗脱(0~10min,12%A;10~18 min,12%A→23%A;18~30 min,23%A→28%A;30~42 min,28%A→45%A;42~70 min,45%A→85%A),流速1.0 mL·min-1,检测波长283 nm,柱温20℃。采用中药色谱指纹图谱相似度评价系统(2004A版)对其进行了相似度计算。结果:建立了毛两面针药材的指纹图谱,并使用聚类分析和主成分分析对指纹图谱进行了模式识别研究。结论:将指纹图谱和模式识别结合起来进行中药材质量控制是一种行之有效的方法,有利于全面、准确的控制药材质量,本研究为评价毛两面针药材质量提供了一定的实验依据。

【Abstract】 Objective: To establish the method of HPLC fingerprint and hierarchical cluster analysis for quality control of Zanthozylum nitidum( Roxb.) DC. f. fastuosum How ex Huang. Methods: The chromatographic fingerprints were obtained by the Promosil C18column( 4. 6 mm × 250 mm,5 μm) with the mobile phase of acetonitrile( A)-water solution( containing 0. 2% phosphoric acid and 0. 2% triethylamine)( B) for gradient elution( 0-10 min,12% A; 10-18 min,12% A→23% A; 18-30 min,23% A→28% A; 30-42 min,28% A→45% A; 42-70 min,45% A→85% A). The wavelength was set at 283 nm and the column temperature was maintained at 20 ℃. The flow rate was 1. 0 mL·min-1. The similarity assay of Z. nitidum f. fastuosum of different sources was carried out to evaluate their quality by traditional Chinese medicine fingerprint similarity evaluation system( 2004A edition). Results: The fingerprint of Z. nitidum f. fastuosum was set up,and cluster analysis and principal component analysis( PCA) were applied to study HPLC fingerprint and chemical pattern recognition. Conclusion: The method can be used for the quality control of Z. nitidum f. fastuosum. The fingerprint and chemical pattern recognition is an effective method for quality control of traditional Chinese medicinal materials.
