【摘要】 目的 探讨PARP-1抑制剂3-AB对脓毒症大鼠心肌细胞PARP-1、Bcl-2表达的影响。方法 成年健康SD大鼠30只,随机分为3组:假手术对照组、模型组和3-AB治疗组。采用盲肠结扎穿孔术制作脓毒症大鼠模型,存活至48h取材。用免疫组织化学SP法和图像分析技术检测心肌PARP-1、Bcl—2的表达,ELISA法检测血清中PARP-1、Bcl-2的含量。结果 模型组心肌PARP-1阳性细胞灰度值、血清中的OD值(optical density)均较假手术组升高,而治疗组较模型组下降,差异有统计学意义,P<0.05;而Bcl-2的变化趋势正好相反,模型组心肌Bcl-2阳性细胞灰度值、血清中的OD值均较假手术组降低,3-AB治疗后其水平较模型组升高,P<0.05。结论 PARP-1抑制剂3-AB可上调脓毒症大鼠大心肌Bcl-2的表达,对凋亡有一定的抑制作用。更多还原

【Abstract】 Objective To explore the effect of 3-AB on expression of PARP-land Bcl-2 in myocardium of rats with sepsis.Methods 30 adult health rats were randomly divided into the sham operation group,the model group,and the therapy group with 3-AB.The ratswere sacrificed on 48 hours after operation.The expression of PARP-1 and Bcl-2 in myocardium was determined by immunohistochemical staining.The immunepositive gray scale of the immune reactant was detected by computer image analysis technique.The OD(optical density)of PARP-1 and Bcl-2 in serum were detected by ELISA.Results In the model group,the expression of PARP-1 in the myocardium increased apparently compared with the sham operation group,P<0.05,but it decreased after therapy with 3-AB,P<0.05.Conversely,the expression of Bcl-2 in the myocardium decreased apparently in the model group after operation compared with the sham operation group,P <0.05,but it increased markedly after therapy with 3-AB compared with the model group,P<0.05.Conclusion The inhibitor of PARP-1(3-AB)could up-regulate the expression of Bcl-2 in myocardium of rats with sepsis and has inhibitive effect of apoptosis.更多还原
