瘦素(leptin)对猪脂肪细胞FTO基因mRNA表达的影响

【摘要】 以体外培养的猪原代脂肪细胞为研究材料,用0,50,100,150 ng/mL瘦素(leptin)分别处理脂肪细胞48 h,油红O染色鉴定脂肪细胞,试剂盒测定细胞中甘油三酯含量,Real-time PCR检测肥胖相关基因(fat mass and obesity associated,FTO)mRNA的表达水平,以探讨不同质量浓度leptin对FTO基因表达的影响。结果表明,猪原代脂肪细胞接种1 d后多数细胞形态呈长梭形或星行,少数呈不规则三角形;5 d细胞加速生长,脂肪细胞开始融合。诱导分化后第3天,可见脂滴被亲脂的油红O着色而呈橘红色。与对照组比较,50 ng/mL,100 ng/mL leptin浓度时细胞内甘油三酯含量和FTO基因的相对表达量显著升高(P<0.05),而150 ng/mL leptin浓度降低细胞内甘油三酯含量和FTO基因的相对表达量。leptin对原代脂肪细胞甘油三酯含量和FTO基因的表达具有浓度依赖性调控作用。

【Abstract】 To investigate the effect of leptin with different mass concentrations on FTO mRNA expression in porcine adipocytes,the adipocytes were treated with 0,50,100,150 ng /mL leptin for 48 h and adipocytes were determined by oil red O,and the content of triglyceride were measured as well asFTO mRNA were detected using real-time PCR technology. The results showed most primary adipocytes were spindle or star shape after 1-day inoculation and cell fusion for 5 days. Third days after differentiation induction,lipid droplets were colored orange and visible with oil red O staining. Compared with the control group,the triglyceride content and the relative expression level of FTO gene increased significantly( P < 0. 05) in 50 and 100 ng /mL leptin concentration,and 150 ng /mL leptin concentration decreased intracellular triglyceride content and the relative expression level of FTO gene. In conclution,leptin regulating the content of triglyceride and the expression level of FTO gene showed concentration dependent in primary adipocyte.
