聚合Cry1C*和VHb基因玉米抗虫耐渍新种质的创制
Creating Maize New Germplasms with Borer-Resistant and Waterlogging-Tolerant by Pyramiding Cry1C* and VHb Gene
【摘要】 分别收集转外源抗虫基因Cry1C*、耐渍基因VHb的HiII、A188玉米材料T1代阳性转化单株花粉,与生产上常用的郑58、248、掖478、昌7-2等30多份自交系杂交和回交,采用"200 mg/L的草铵膦初选-PCR检测-表型鉴定"筛选程序对后代植株进行阳性单株筛选,连续进行3次回交和筛选得到BC3家系,将具有相同遗传背景、不同目标基因的家系杂交,随后选择含2个目标基因的个体与轮回亲本回交和筛选,至BC5后自交纯合获得双抗新系11份。所得到的新系在抗虫、耐渍性状上与轮回亲本有显著差异。
【Abstract】 The pollens from positive plants of T1 generation of Hi II and A188, which were transformed with waterlogging tolerance gene"VHb"and borer-resistant gene"Cry1C*"through Agrobaterium-mediated transformation system and particle gun bombardment, respectively, and were collected to pollinate more than 30 important inbred lines such as Zheng58, 248, Ye478, Chang7-2 and so on. And then backcross with these inbred lines as recurrent parents. Subsequently, positive plants were obtained by performing"200 mg/L phosphinothricin screening-PCR validation-phenotype identification"program. The BC3families were obtained after backcrossing and screening for 3 times continuously, and then the families with same genetic background but different target genes were crossed to gain those individuals possessing two target genes. Further, the above-mentioned individuals were backcrossed with the corresponding recurrent parents, and then the progenies were screened to get goal plants. Eventually, after continuous backcrossing and screening for 5 times, 11 new double-resistant lines containing "Cry1C*"and "VHb"gene were obtained by inbreeding. There were significant differences between the new lines and the recurrent parent in the traits of borer resistance and waterlogging tolerance.
