高蛋白饮食对小鼠运动能力和内质网应激的影响
Enhancement of Endoplasmic Reticulum Stress and Reduction of Exercise Capacity of Mice Caused by High Protein Diet
【摘要】 目的:探讨高蛋白饮食对小鼠运动能力和内质网应激的影响。方法:雄性C57/BL6小鼠随机分为对照组和高蛋白组,连续3天给予能量相同的饮食,各组又分为对照安静组、对照运动组、高蛋白安静组和高蛋白运动组。运动组进行一次性力竭游泳。4小时后取标本,测定肝脏和股四头肌内质网应激蛋白及mRNA的表达量,测定血糖、肝糖原和肌糖原水平。结果:与对照运动组相比,高蛋白运动组游泳至力竭时间显著缩短(P<0.01),肝脏和股四头肌内质网应激标志性蛋白GRP78(P<0.05)、p-IRE1/IRE1和GRP78、XBP1s、CHOP和GADD34的mRNA表达显著增加(P<0.01),血糖水平各组间无显著性差异。与对照安静组相比,高蛋白安静组肝糖原和肌糖原储备均显著下降(P<0.05,P<0.01)。结论:高蛋白饮食缩短运动持续时间,并可引起内质网应激增强。
【Abstract】 Objective To observe the effects of high protein diet on the endoplasmic reticulum stress(ERS)in liver and skeletal muscle,and the exercise capacity of mice.Methods C57/BL6 male mice were randomly divided into control group and high protein diet group(HP).They were respectively given same total energy of normal diet or high protein diet for 3 days,and then separated into control sedentary group(C),control exercise group(CE),high protein diet sedentary group(HP)and high protein diet exercise group(HPE).The exercise groups underwent an acute exhaustive swimming,and the time to exhaustion was recorded,the expression of protein and mRNA of ERS biomarkers in liver and quadriceps were measured 4 hours later,the contents of blood glucose and glycogen in liver and muscle were assayed.Results The time to exhaustion in group HPE was shorter than in group CE(P < 0.01).Compared to the group CE,the protein expression of GRP78 and p-IRE1/IRE1,and the mRNA level of GRP78,XBP1 s,CHOP,and GADD34 in liver and quadriceps increased markedly in group HPE(P <0.05 and P < 0.01).There was no significant difference in the concentration of blood glucose between groups CE and HPE.Compared to the group C,glycogen in liver and muscle decreased significantly in group HP(P < 0.05 and P < 0.01),while the expression of protein and mRNA of ERS biomarkers increased slightly without statistical difference.Conclusion High protein diet reduces the duration of exercise and enhances the degree of endoplasmic reticulum stress in mice.
