银丝草对四氯化碳诱导小鼠急性肝损伤的保护作用

【摘要】 目的观察银丝草对四氯化碳(CCl4)所致小鼠急性肝损伤的保护作用。方法将60只昆明种小鼠随机分成6组:正常对照组、模型组、阳性对照组(联苯双酯滴丸)、银丝草低、中、高剂量组(10 g生药/kg、20 g生药/kg、40 g生药/kg),每组10只。通过皮下注射0.2%CCl4花生油溶液建立小鼠急性肝损伤模型,测定小鼠血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)的活性及肝组织中丙二醛(MDA)的含量和超氧化物歧化酶(SOD)的活力;计算肝脏指数并同时对肝组织进行病理学检查。结果银丝草高、中、低剂量组都能不同程度地降低CCl4致小鼠急性肝损伤血清ALT,AST活性,降低肝匀浆中MDA的含量,增强SOD的活力(P<0.05)。通过病理学切片观察,银丝草各剂量组能明显改善肝组织的病理变化。结论银丝草对四氯化碳引起的小鼠急性肝损伤具有显著的保护作用,其作用机制可能与清除体内自由基和抗氧化有关。

【Abstract】 Objective To observe the protective effect of aqueous extract of Evolvulus alsinoides on acute liver injury induced by CCl4 in mice. Methods Sixty mice were randomly divided into 6 groups as follows: normal control group,model group,the positive control group( Bifendate),low,medium and high-dose groups of aqueous extract of Evolvulus alsinoides( 10,20,40 g /kg),10 rats per group,all groups except the normal control were subcutaneous injected with0. 2% CCl4 solution to induce acute hepatic injury. The level of alanine transaminase( ALT),aspartate transaminase( AST) in serum and malondiadeyde( MDA) content,the activity of superoxide dismutase( SOD) in liver were determined. In addition,the liver pathological examination and the changes of mice liver index were observed. Results Compared with model group,the level of ALT and AST in serum of each treatment group were significantly decreased( P< 0. 05),the content of MDA in the liver was also significantly lowered( P < 0. 05),and the activity of SOD in liver was significantly improved after preventive treatment with aqueous extract of Evolvulus alsinoides. Conclusion Evolvulus alsinoides has protective effect on CCl4-induced acute liver injury in mice,and the mechanism is probably related with its scavenging free radical and antioxidant activites.
