负载As2O3壳聚糖纳米粒对大鼠免疫功能的影响

【摘要】 目的 观察负载三氧化二砷(arsenic trioxide,As203)壳聚糖纳米粒(NPCS-As203)对大鼠免疫功能的影响,为临床用药提供实验和理论依据。方法 将40只SD大鼠随机分为4组:壳聚糖纳米粒组(NPCS组),负载As203壳聚糖纳米粒组(NPCS-As2O3组),As2O3组和生理盐水(NS)组。尾静脉注射给药,连用7天后处死大鼠,取脾称重计算脾脏指数并分离脾细胞进行特异性细胞毒T淋巴细胞(CTL)功能检测。结果As2O3组CTL活性降低,与NS组相比,差异有统计学意义(P<0.05);NPCS组和NPCS-As2O3组均显示出较高的CTL活性,并且脾脏指数增加,与As2O3组和NS组比较差异均有统计学意义(P<0.05)。结论 As2O3可以降低大鼠脾免疫功能;NPCS可以明显提高大鼠脾免疫功能;NPCS-As2O3可以减轻As2O3造成的脾免疫功能损伤,保护大鼠脾的免疫功能。

【Abstract】 Objective To explore the effects of As2O3 loaded chitosan nanoparticle(NPCS-As2O3) on the immune function in rats,which can provide experimental and theoretical evidence for better clinical therapy.Methods Forty SD rats were randomly divided into four groups:a chitosan nanoparticle(NPCS) group,an As2O3 loaded chitosan nanoparticle(NPCS-As2O3) group,an As2O3 group and a normal saline(NS) group.The agents were given via tail vein injection for seven days.The spleens were removed to dertermine spleen index,while spleen cells were separated to test the function of specific cytotoxic T lymphocytes(CTLs).Results The activity of CTL in the As2O3 group was remarkably reduced when compared with that in the control group(P <0.05).Both tthe NPCS group and the NPCSAs2O3 group presented stronger activities of CTL and higher values of spleen index than those in the As2O3 group and NS group(P <0.05).Conclusions The immune function in rats can be damaged by As2O3 but remarkably enhanced by NPCS.NPCS-As2O3 could relieve the immune damage caused by As2O3.The presence of NPCS-As2O3 suggests a feasible idea of reducing the toxic and side-effects of As2O3 on clinical tumor therapy.
