工频电磁场对作业人员淋巴细胞凋亡相关基因mRNA水平影响研究
Expression levels of lymphocyte apoptosis-related genes in power-frequency electromagnetic field operation personnel
【摘要】 目的探讨工频电磁场对作业人员周围血淋巴细胞凋亡相关基因P53、B细胞淋巴瘤/白血病基因2(Bcl-2)及其相关X蛋白基因(Bax)的mRNA水平及血浆天冬氨酸特异性半胱氨酸蛋白酶3(Caspase-3)蛋白水平的影响。方法随机选择某500 kV变电站工龄5年以上的45名工频电磁场巡检作业工人作为接触组,并按照1∶1配对选取该变电站45名行政管理及后勤人员作为对照组。2组研究对象均采集周围静脉血,提取淋巴细胞,采用实时荧光定量聚合酶链式反应测定其淋巴细胞中P53、Bcl-2和Bax的mRNA水平,采用酶联免疫吸附实验法测定血浆Caspase-3蛋白表达水平。数据采用中位数与四分位数间距[M(P25,P75)]描述。结果接触组和对照组人群P53mRNA相对表达量(RQ)分别为4.00(3.60,4.37)和4.21(3.70,4.64),Bcl-2 mRNA RQ为4.07(3.47,4.50)和4.04(3.48,4.35),Bax mRNA RQ为4.37(4.02,4.89)和4.53(4.11,4.85),Bcl-2与Bax的mRNA RQ比值为0.89(0.81,1.01)和0.87(0.80,0.95),Caspase-3蛋白表达水平分别为7.56(4.05,15.32)和9.70(6.40,21.13)pmol/L。2组上述5个指标比较,差异均无统计学意义(P>0.05)。结论长期接触工频电磁场对机体淋巴细胞凋亡相关基因P53、Bcl-2、Bax的mRNA水平及血浆Caspase-3蛋白水平影响不明显。
【Abstract】 Objective To explore the effect of the power-frequency electromagnetic field on lymphocyte apoptosis related genes P53,B-cell lymphoma / Leukemia-2( Bcl-2),Bcl-2-associated X protein( Bax) mRNA level and the peripheral blood plasma cysteine-containing aspartate-specific proteases 3(Caspase-3) protein concentration in the workers. Methods Forty-five workers with length of service more than 5 years working in a 500 kV substation were randomly chosen as the exposed group,and in accordance with the age,length of service as a match we selected the corresponding percentage(1∶ 1) of substation administrative and support staff of 45 as the control group. Respectively,lymphocytes from 10 ml peripheral blood were extracted,real-time polymerase chain reaction was used to determine P53,Bcl-2 and Bax mRNA level in the lymphocyte,and enzyme linked immunosorbent assay was used to test peripheral blood plasma Caspase-3 protein concentration. Results In the exposed group and control group,P53 mRNA expression of relative quantity( RQ) were 4. 00(3. 60,4. 37) and 4. 21(3. 70,4. 64),Bcl-2 mRNA RQ were 4. 07(3. 47,4. 50) and 4. 04(3. 48,4. 35),Bax mRNA RQ were 4. 37(4. 02,4. 89) and 4. 53(4. 11,4. 85),Bcl-2 mRNA RQ / Bax mRNA RQ were 0. 89(0. 81,1. 01) and 0. 87(0. 80,0. 95),besides Caspase-3 protein concentration were 7. 56(4. 05,15. 32) and 9. 70(6. 40,21. 13) pmol / L. The above indicators between the 2 groups had no statistical significance differences( P > 0. 05). Conclusion The effect of power-frequency electromagnetic field exposure on the lymphocyte apoptosis related genes P53,Bcl-2,Bax mRNA level and peripheral blood plasma Caspase-3 protein concentration was not obvious in the current study.
