【摘要】 目的研究血管内皮生长因子(VEGF)蛋白及其mRNA在乳腺癌中的表达,探讨其在乳腺癌发生、发展过程中的临床意义。方法运用免疫组化检测2010年1月至2012年12月56例乳腺癌和30例乳腺纤维腺瘤组织(良性病变组织)中VEGF蛋白水平,采用半定量反转录聚合酶链反应(RT-PCR)方法检测mRNA的表达情况。结果 VEGF蛋白在乳腺癌组织中阳性表达率为75.0%(42/56),在乳腺良性病变组织中为23.3%(7/30),二者比较,差异有统计学意义(P<0.05);VEGF mRNA在乳腺癌和乳腺良性病变组织中相对表达量分别为0.812 7±0.104 2、0.327 5±0.038 4,二者比较,差异有统计学意义(P<0.05);VEGF蛋白及其mRNA在乳腺癌中的表达与淋巴结转移呈正相关。结论 VEGF蛋白及其mRNA有可能作为判断乳腺癌浸润转移能力的参考指标之一。

【Abstract】 Objective To study the expression of vascular endothelial growth factor(VEGF) protein and its mRNA in breast cancer and evaluate its clinical significance in the occurrence and development of breast cancer. Methods Immunohistochemistry was used on 56 patients with breast cancer and 30 patients with breast fibroadenoma tissues(benign lesion) from January 2010 to December 2012 to detect the level of VEGF protein,and semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) was adopted to detect the expression of mRNA. Results The positive expression rate of VEGF protein was 75.0%(42/ 56) in breast cancer tissue and 23.3%(7/30) in benign breast tissue,and the difference had statistical significance(P<0.05). The relative expression of VEGF mRNA in breast caner tissue and benign breast tissue was 0.812 7 ±0.104 2 and 0.327 5±0.038 4 respectively,which showed statistically significant difference(P<0.05). The VEGF protein and mRNA expression in human breast cancers were positive correlated with lymphatic metastasis. Conclusion The VEGF protein and its mRNA can be a predictor for the ability of breast cancer invasion and metastasis.
