快速测定石斛多糖含量方法的优化

【摘要】 通过取样量、超声时间、浸提剂量、粒径等4个石斛多糖的超声提取影响因素的正交和单因素试验。结果表明,石斛多糖含量超声提取测定的最佳参数为25.0 mg的取样量、80目的粒径、40 min的超声时间、浸提试剂体积与取样量之比为220。该方法具有样品前期处理时间短,测定仪器操作简便,测定速度快、样品用量少的特点,能在较短时间内对大量单株个体进行检测鉴定,为石斛的品质育种提供了有效途径和手段。

【Abstract】 Four factors including sampling quantity,treating time,volume of extracting solution and granule size influencing the extract by ultrasonic were researched synthetically. The results were determined on the basis of orthogonal experiment and one-factor tests. The best parameters was collected for the extraction of the polysaccharides,sampling quantity was 25. 0 mg crushed to 80 eye,treating time was 40 min,the ratio of power to water was 1∶ 220. The advantages of this methods including short method could be applied to determine the polysaccharides content of plentiful plants in a short time and the genotype individual possessing of high content of polysaccharides may be screened out,which could be used as the effective measure in the quality-meliorating of Dendrobium officinale.
