新疆巴什拜羊BPI基因序列的生物信息学分析

【摘要】 以新疆巴什拜羊BPI基因为研究对象,运用生物信息学方法对其编码蛋白的结构、理化性质、信号肽、跨膜结构、亚细胞定位、二级结构以及高级结构进行生物信息学分析,并推测与其他物种的生物进化关系。结果表明,BPI蛋白分子量为53.4 ku,理论等电点大于7,呈碱性,N端有信号肽,肽链表现为亲水性,基本属于分泌蛋白。BPI蛋白质的二级结构为α螺旋、β折叠和无规则卷曲,有2个保守结构域BPI1和BPI2。通过进化树分析发现巴什拜羊的BPI基因在绵羊、牛、虎鲸、野猪、人、猕猴、家兔、小鼠、非洲爪蟾中,与绵羊首先聚为一类,后与牛聚为一类,这与动物学分类结果一致,这种同源性在一定程度上代表着物种亲缘关系的远近。

【Abstract】 The bactericidal permeability increasing protein(BPI) gene in Bashibai sheep was investigated by bioinformatics analysis.Its encoding protein was analyzed including its physical and chemical property,signal peptide,transmembrane structure,subcellular localization,secondary structure and high-level structure,and speculated its evolution relationship with other species.The results showed that the BPI protein of 53.4ku,the theoretical isoelectric point greater than 7,alkaline,N-terminal signal peptide,the peptide chain perform hydrophilice,the basic part and the transmembrane protein secreted proteins.BPI protein secondary structure for α-helics,β-strands and random coil,contained two conserved domains BPI1 and BPI2.In conclusion BPI gene of Bashibai sheep has closest relationship with that of ovis aries,compared with that of ovis aries,bos taurus,orcinus orca,sus scrofa,homo sapiens and oryctolagus based on the analysis of molecular evolution.
