高致病性猪繁殖与呼吸综合征病毒F株的分离鉴定

【摘要】 2012年2月,潍坊某猪场疑似发生高致病性猪繁殖与呼吸综合征(PRRS)。病猪主要表现为呼吸困难,耳朵发绀,皮肤有弥漫性出血点;剖检发现淋巴结、肺脏出血严重。从病死仔猪脾、肺、淋巴结和脑组织中分离得到病毒。将该毒株在Marc-145细胞上盲传3代出现典型细胞病变,F3代病毒滴度为106.5TCID50/mL。RT-PCR检测结果显示,猪繁殖与呼吸综合征病毒(PRRSV)阳性。用F3代毒株进行动物回归试验,结果符合高致病性PRRSV致病特征。上述结果证明该猪场发生的传染病为高致病性PRRS,并且利用Marc-145细胞成功分离到高致病性PRRSV,命名为F株。本研究为高致病性PRRSV致病特点及其疫苗研制提供物质基础。

【Abstract】 In February,2012,the suspected cases of highly pathogenic porcine reproductive and respiratory syndrome( PRRS) were detected in Weifang. The main manifestations of diseased pigs were dyspnea and cyanosis of the ear and vulva; necropsy revealed serious hemorrhage in lymph node and lung. A strain of virus was isolated from spleen,lung,lymph node and brain tissues in infected piglets. Cytopathic effect was obvious following three blind passages on Marc- 145 cells,and the virus titer for the third generation of the virus was106. 5TCID50/mL. The isolate was demonstrated to be porcine reproductive and respiratory syndrome virus( PRRSV) by RT-PCR. The results of animal challenge test showed its characteristics of highly pathogenic PRRSV. The isolated virus was named F strain. This study provides the material basis for the investigation of pathogenic characteristics of PRRSV and vaccine development.
