汶上芦花鸡种质资源的遗传多样性分析
Analysis on the Genetic Diversity of Wenshang Barred Chicken Breed
【摘要】 为了评估汶上芦花鸡群体的遗传多样性,本研究采用联合国粮农组织(FAO)和国际动物遗传学会(ISAG)联合推荐的27对微卫星引物,对济宁汶上芦花鸡的100个个体(60只母鸡,40只公鸡)进行了遗传多样性检测,分析了该群体在DNA水平上的遗传多样性。结果,共检测到86个等位基因,每个微卫星座位的等位基因数从2个到6个不等,平均等位基因数为3.185,平均期望杂合度和多态信息含量分别为0.465 5和0.402 3。对分布在不同地方的3个群体的遗传距离分析表明,群体间的遗传距离为0.05~0.10。以上结果表明,汶上芦花鸡群体的遗传多样性偏低,必须采取一定的措施保护这一优良地方品种。
【Abstract】 In this study,one hundred chickens including 60 hens and 40 roosters were sampled for the evaluation of genetic diversity on Wenshang Barred chicken breed by using 27 pairs of primers recommended jointly by FAO(Food and Agriculture)and ISAG(the International Society for Animal Genetic).The genetic diversity at DNA level were analyzed in this study.Totally 86 alleles were detected,the number of alleles varied from 2to 6,giving a mean value of 3.185 alleles perlocus.The average heterozygosity and polymorphism information content of 27 microsatellite loci were 0.465 5and 0.402 3,respectively.The genetic distances between 3populations distributed in different areas vary from 0.05 to 0.10.The results indicated that Wenshang Barred chicken breed had low heterozygosity and polymorphism information content,which imply that effective methods should be taken to protect this breed.
