【摘要】 目的研究阿司匹林(aspirin)对宫颈癌Siha细胞凋亡和增殖的影响。方法培养人宫颈癌Siha细胞,加入不同浓度阿司匹林(1.0 mmol/L、5.0 mmol/L、10.0 mmol/L)干预培养,HE染色观察Transwell小室穿梭能力;划痕愈合实验观察迁移能力;应用流式细胞技术检测Siha细胞凋亡和细胞周期的变化;TUNEL染色检测细胞凋亡情况;用RT-PCR检测细胞凋亡相关基因bcl-2及bax转录水平的改变;蛋白质印迹法检测阿司匹林对宫颈癌Siha细胞bcl-2及bax蛋白表达的影响。结果阿司匹林表现出明显地抑制人宫颈癌Siha细胞的迁移、增殖以及促进凋亡(P<0.05),且伴随浓度的递增和时间的延长,抑制作用更明显。结论阿司匹林具有较强的抗入宫颈癌作用,其机制可能与阿司匹林诱导宫颈癌细胞凋亡,并抑制其增殖有关。更多还原

【Abstract】 Objective To assess the effects of aspirin on the proliferation and apoptosis of human cervical cancer Siha cell line.Methods Siha cell was cultured and induced with aspirin of concentration gradient(1.0mmol/L,5.0mmol/L,10.0mmol/L).H.E staining was used to observe Transwell chamber shuttle capacity;scratch healing experiment observed migration;the effect of aspirin on Siha cell apoptosis and cell cycle was observed by cell flow cytometry(FCM),and the apoptosis was shown with TUNEL staining.The levels of bcl-2 and bax mRNA were detected by RT-PC.Expression of bcl-2 and bax protein was determined by Western blotting.Results The results showed aspirin significantly inhibited migration,proliferation of human cervical cancer Siha cell and promoted its apoptosis(P<0.05),and with increasing concentration and time extension,the trend was more obvious.Conclusion Aspirin has strong inhibition effects on cervical cancer,its mechanism may be related to induction of apoptosis on cervcal cancer,and inhibition of proliferation.更多还原
