钙对氟致大鼠睾丸损伤的拮抗作用研究

【摘要】 目的研究氟致大鼠睾丸组织的损伤作用以及钙的拮抗作用。方法选用健康雄性清洁级SD大鼠80只,随机分为10组:对照组、加钙染氟6个剂量组(NaF 2.1、4.2、8.5、17、34、68 mg/kg·d;CaCO325、50、50、100、100、150 mg/kg·d)和单纯染氟3个剂量组(NaF 2.1、8.5、34 mg/kg·d),灌胃染毒90 d,染毒结束次日,观察氟、钙对大鼠睾丸脏器系数、睾丸结构的影响,并用氟离子选择电极法检测睾丸氟含量,用原子吸收分光光度计测定睾丸中钙(Ca)、铁(Fe)和锌(Zn)元素水平。结果氟可使大鼠睾丸脏器系数降低并导致睾丸病理学改变。各组大鼠睾丸氟含量组间比较,差异有统计学意义(F=62.224,P=0.000)。表现为加钙染氟第4、5、6组和高、中、低剂量染氟组睾丸氟含量均高于对照组。高、中剂量染氟浓度下加钙组睾丸氟含量低于单纯染氟组。各组大鼠睾丸Ca、Fe和Zn水平组间比较,差异有统计学意义(F值分别为7.658、6.177、10.802,P<0.05),表现为高、中、低剂量染氟组Ca、Fe和Zn水平均低于对照组,加钙组睾丸Ca、Fe和Zn水平高于高剂量染氟组。结论氟可在睾丸内蓄积、破坏睾丸结构并改变睾丸中一些金属元素水平,适量的钙可拮抗氟致大鼠睾丸组织的损害作用。

【Abstract】 Objective The study was aimed to discuss the effects of fluoride in rat testis and the antagonistic effect of calcium on testis injury induced by fluoride. Methods A total of 80 healthy male SD rats were equally randomized into ten groups: control group, six fluorosis groups with calcium(NaF 2.1, 4.2, 8.5, 17, 34, 68mg/kg·d; CaCO325, 50, 50, 100, 100, 150 mg/kg·d) and three fluorosis groups(NaF 2.1, 8.5, 34mg/kg·d). The solution of sodium fluoride and calcium carbonate were administered to rats by intragastric administration for 90 days. After the last dosing, the organ coefficients of testis and the pathological changes were monitored. The concentrations of fluoride in testis were examined with F-ion selective electrode. The concentrations of calcium(Ca), iron(Fe) and zinc(Zn) were measured by atomic absorption spectrophotometer. Results Fluoride could reduce the organ coefficients and damage the structure of testis. The differences of fluoride concentration in testis among groups were statistically significant(F=62.224, P=0.000). The concentration of fluoride in group 4, group 5, and group 6 as well as in high, middle, and low fluorosis groups were significant higher than the control group. The concentration of fluoride in calcium groups were significant lower than the high and middle fluorosis groups. The difference of Ca, Fe and Zn levels among groups were statistically significant(F=7.658, 6.177, 10.802, P<0.05). The levels of Ca, Fe and Zn in three fluorosis groups were lower than the control group. The levels of Ca, Fe and Zn in calcium groups were significant higher than the high fluorosis group. Conclusion Fluoride can accumulate in testis, damage the structure of testis and affect the levels of metal elements. Appropriate level of calcium could antagonize the testis injury induced by fluoride.
