合肥市W135群脑膜炎奈瑟菌的分子分型研究

【摘要】 目的利用脉冲场凝胶电泳技术(pulsed-field gel electrophoresis,PFGE)对合肥地区2012年分离到的W135群脑膜炎奈瑟菌进行分型,探讨其同源性。方法 1株疑似流脑病例的血液及3个疑似病例的密切接触者咽拭子进行脑膜炎奈瑟菌的培养分离,对分离的菌株进行生化试验、血清学分群鉴定、PCR扩增和PFGE鉴定试验。结果合肥地区分离出11株W135群脑膜炎奈瑟菌,11株W135群脑膜炎奈瑟菌经PFGE分子分型可以分为合肥1到合肥5型,分别是合肥1型4株,合肥2型1株,合肥3型2株,合肥4型1株和合肥5型3株。结论合肥地区的11株W135群脑膜炎奈瑟菌分为5个基因型。

【Abstract】 Objective This study aimed to investigate the homology of the W135 Group of Neisseria meningitidis isolated in Hefei area in 2012 by pulsed-field gel electrophoresis(PFGE). Methods The blood sample of a suspected epidemic meningitis patient and throat swabs of three close contacts were collected for pathogen identification. Biochemical reaction analysis, serotyping, PCR amplification and PFGE were carried out to identify Neisseria meningitidis from the isolated bacteria. Results 11 isolates from Hefei area were identified as Neisseria meningitides of serotype W135 by PFGE. The distribution of their subtypes was 4 of HF1, 1 of HF2, 3 of HF3; 1 of HF4 and 3 of HF5. Conclusion 11 Neisseria meningitides of serotype W135 were identified by PFGE from isolates of Hefei area, and further divided into five genotypes of HF1, HF2, HF3, HF4 and HF5.
