甲异靛诱导K562细胞凋亡及对JAK/STAT途径效应基因表达影响的研究

【摘要】 目的探讨甲异靛诱导K562原代细胞凋亡及对效应基因表达影响的作用。方法 K562细胞甲异靛处理12 h、24 h、48 h后,采用流式细胞术检测细胞凋亡情况,Western blot检测Bcl-xL、CyclinD1和C-MYC的改变。结果甲异靛作用K562细胞12 h,24 h,48 h组的凋亡率与对照组比较有显著性差异(P均<0.05);K562组24 h、48 h与12 h比较有显著性差异(P均<0.05)。在甲异靛作用12~48 h,C-MYC蛋白含量逐渐减少,而Bcl-xL、CyclinD1无显著变化。结论甲异靛促进K562细胞凋亡,可能与下调STAT5下游C-MYC的蛋白含量表达减少有关。

【Abstract】 Objective It is to investigate the effect of Meisoindigo induce primary K562 cell apoptosis and its influence on the expression of corresponding gene. Methods K562 cell was treated with Meisoindigo for 12 h,24 h,48 h respectively,flow cytometry was used to detect cell apoptosis,Western blot was employed to examine the alteration of the expression of protein Bcl- xL,Cyclin D1 and C- MYC. Results Compared with the control group,the differences in the apoptosis rates of K562cell treated with Meisoindigo for 12 h,24 h,48 h were significant( P < 0. 05),the differences in the rates were also significant between 12 h group and 24 h group or 48 h group. After the treatment of Meisoindigo for 12 h to 48 h,the expression of C-MYC was decreased gradually,while Bcl- xL,Cylin D1 exhibited no significant difference. Conclusion Meisoindigo can accelerrate the apoptosis of K562 cell and the mechanism may relevant to the decreased expression of C- MYC which located downstream of STAT5.
