经皮下注射CCl4结合饮用乙醇致大鼠肝硬化模型的研究

【摘要】 目的本实验通过对大鼠经皮下注射CCl4结合饮用乙醇制作肝硬化模型,观察肝脏显微结构、超微结构及血清学指标的变化,以期为今后更有效地研究肝硬化提供依据。方法健康雄性Wistar大鼠模型组、对照组各10只,模型组大鼠经皮下注射CCl4结合饮用乙醇14周,应用HE染色和电镜技术观察肝脏显微结构和超微结构变化并对血液进行血清学指标LN、CⅣ、PCⅢ、HA的检测。结果与对照组相比,模型组显微及超微结构出现了肝硬化的表现,血清学指标提示肝纤维化明显。讨论对大鼠经皮下注射CCl4结合饮用乙醇可以成功制作肝硬化模型。

【Abstract】 Objective It is to observe the changes of microstructure and ultrastructure and analyze the changes of serologic index in the rat liver cirrhosis model established by subcutaneous injection of CCl 4 combining with drinking ethonal in order to provide evidences for studying liver cirrhosis more effectively in future. Methods The healthy male Wistar rats were divided into model and control groups( n = 10). Within 14 weeks,model group was subcutaneously injected by CCl 4 combining with drinking ethonal. The changes of microstructure and ultrastructure in liver were observed by HE staining and technique of electron microscope and the serum levels of LN,CⅣ,PCⅢ,HA were determined. Results Compared with the control group, the microstructure and ultrastructure of liver cirrhosis and hepatic fibrosis could be found obviously in model group. The serum indexes also indicate there was a obvious hepatic fibrosis. Conclusion The rat liver cirrhosis model can be successfully established by subcutaneous injection of CCl 4 combining with drinking ethonal.
