黄荆子乙酸乙酯提取物对人乳腺癌MCF-7细胞Mcl-1、Bcl-2、Bax蛋白的影响

【摘要】 目的研究黄荆子乙酸乙酯提取物(EVn-50)对人乳腺癌MCF-7细胞Mcl-1、Bcl-2、Bax蛋白的作用。方法应用Wersten Bloting分析EVn-50对人乳腺癌MCF-7细胞Mcl-1、Bax和Bcl-2蛋白表达的影响。结果 Evn-50使Mcl-1及Bcl-2值下调,Bax值上调,促进细胞凋亡。且EVn-50中浓度组相对灰度面积比值与低浓度组比较有显著性差异(P均<0.05),EVn-50组48 h相对灰度面积比值与24 h比较有显著性差异(P均<0.05)。结论 EVn-50具有诱导人乳腺癌MCF-7细胞凋亡的作用,最佳浓度为3.0 mg/L,最佳作用时间为48 h。

【Abstract】 Objective It is to observe the effect of the acetoacetate extract of the Vitex negundo seeds( EVn-50) on the human breast cancer MCF-7 cell Mcl-1,Bcl-2 and Bax protein. Methods The acetoacetate extract of the Vitex negundo seeds on human breast cancer MCF-7 cell Mcl-1,Bcl-2 and Bax protein expression were detected by Western Bloting analysis. Results Evn-50 made the expression of Mcl-1 and Bcl-2 downward,and made the expression of Bax increased, thereby promoted cell apoptosis. There were significant in the relative concentration area ratio between EVn-50 mid-concentration group and Low-concentration group( all P < 0. 05); and there were significant difference between treated for 48h and 24h( all P < 0. 05) 。Conclusion Evn-50 can induce the apoptosis of human breast cancer MCF-7 cells. The optimal concentration was 3. 0 g,and the best time is 48 hours.
