哈茨木霉发酵生产MonocillinⅠ的培养基优化研究

【摘要】 采用响应面分析法优化哈茨木霉菌发酵生产MonocillinⅠ培养基。以monocillinⅠ产量为指示,通过HPLC/MS测定其产量。在单因素试验基础上,选定麦芽糖、黄豆粉和磷酸二氢钾添加量3个因素做中心组合试验,建立二次回归方程,并用响应面分析法进行优化。结果表明:绿色木霉菌的最优碳源、氮源、磷源及其最优添加量分别为麦芽糖4.162%、黄豆粉0.253%、磷酸二氢钾0.044 5%。在此条件下培养的哈茨木霉菌发酵产生MonocillinⅠ产量达到16.241mg/L,表明响应面分析法非常适合优化MonocillinⅠ发酵培养基,使产量提高了34.5%。因此,培养基优化方法高效、简便、耗时少,为MonocillinⅠ发酵奠定工业基础。

【Abstract】 Response surface methodology(RSM)was adopted to optimize the media for culturing monocillin Ⅰby Trichoderma harzianum88.MonocillinⅠyield was determined by HPLC/MS.According to the single-factor experiments,three factors(amount of maltose,soyflour and KH2PO4)were selected for Box-Behnken designed to establish a quadric regression equation.The optimal carbon,nitrogen,and phosphorus resources and their adding amounts were established as maltose(4.162%),soyflour(0.253%),and KH2PO4(0.044 5%).Under the optimal conditions,ayield of 16.24mg/L was achieved,increased by 34.5%.The chosen method of medium optimization was efficient,simple and less time consuming,which laid a foundation of large-scale production MonocillinⅠ.
