新生儿缺氧缺血性脑病患儿血液中血管内皮生长因子和miR-125b表达水平及相关性研究

【摘要】 目的:探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)和miR-125b在新生儿缺氧缺血性脑病(hypoxic-ischemic encephalopathy,HIE)患儿和正常新生儿血液中表达水平及相关性。方法:收集20例正常新生儿和20例HIE患儿血液标本,分离血浆和外周血单个核细胞(peripheral blood mononuclear cell,PBMC),利用real-time PCR检测PBMC中miR-125b表达,ELISA方法检测血浆中VEGF含量。结果:与正常对照组相比,HIE患儿PBMC中miR-125b表达显著降低(P=0.00002),而血浆中VEGF水平显著升高(P=6.26×10–8);同时在正常组中,miR-125b表达与VEGF水平无相关性(r=0.016,P=0.946,n=20),但在HIE组(r=–0.505,P=0.023,n=20)二者却呈显著负相关。结论:新生儿脑缺血缺氧时可能通过降低miR-125b表达从而升高VEGF表达。

【Abstract】 Objective:To explore the expression and correlation between vascular endothelial growth factor(VEGF) and miR-125 b in healthy and hypoxic-ischemic encephalopathy(HIE) neonatus.Methods:A total of 20 controls and 20 HIE samples were recruited in this study,plasma and peripheral blood mononuclear cell(PBMC) were prepared for detecting the miR-125 b expression and VEGF concentration using realtime PCR and ELISA kit,respectively.Result:Compared with controls,miR-125 b expression in PBMC was significantly decreased(P=0.00002) but VEGF concentration in plasma was increased in HIE group(P=6.26×10–8).Meanwhile,there was no significant correlation between miR-125 b expression and VEGF level in controls(r=0.016,P=0.946,n=20).However,significant correlation was observed in HIE group(r=–0.505,P=0.023,n=20).Conclusion:The VEGF level was increased possibly due to the downregulation of miR-125 b in HIE neonatus.
