三甲散对HSC-T6细胞α-SMA、TIMP-1和ROCK-Ⅰ蛋白表达的影响
Effect of Sanjia powder on expression of protein of α-SMA, TIMP-1, ROCK-Ⅰ in hepatic stellate cell
【摘要】 [目的]检测三甲散含药血清对肝星状细胞株(HSC-T6)细胞中α-平滑肌肌动蛋白(α-SMA)、基质金属蛋白酶抑制-1(TIMP-1)和蛋白激酶(ROCK-Ⅰ)的表达,从而研究三甲散对HSC-T6细胞增殖作用的影响。[方法]以三甲散含药血清干预HSC-T6细胞,采用实时荧光定量聚合酶链式反应(RT-PCR)和免疫印迹(Western blot)技术,检测三甲散含药血清对HSC-T6细胞中α-SMA、TIMP-1和ROCK-Ⅰ的蛋白表达。[结果]三甲散和Y-27632显著地降低了HSC-T6细胞α-SMA、ROCK-Ⅰ和TIMP-1 m RNA和蛋白的表达。[结论]三甲散可降低HSC-T6细胞ROCK-Ⅰ蛋白、TIMP-1和α-SMA的表达,从而抑制HSC-T6细胞的增殖。
【Abstract】 [Objective] The expression of α-SMA, TIMP-1 and ROCKⅠ in the hepatic(HSC-T6)stellate cell were measured to study the proliferating effect of Sanjia powder. [Methods] HSC-T6 cells interfered by Sanjia powder-containing serum, RT-PCR and Western blot were used to detect the expression of α-SMA, TIMP-1, ROCKⅠ in the hepatic stellate cell. [Results] Real-time PCR and Western blot assay showed that Sanjia powder and Y-27632 could reduce the expression of ROCK-Ⅰprotein, TIMP-1 and a-SMA m RNA in HSC-T6 cells. [Conclusion]Sanjia powder can decrease the expression of ROCK-Ⅰ protein, TIMP-1 and a- SMA in HSC-T6 cells.
