1,25-二羟维生素D3对严重烫伤小鼠应激反应的影响
Effect of 1,25-Dihydroxyvitamin D3 on Alleviating the Stress Response in Severely Scalded Mice
【摘要】 目的探讨1,25-二羟维生素D3[1,25-(OH)2VitD3]对严重烫伤小鼠应激状态下胰岛素抵抗和炎症反应的影响。方法 130只小鼠按随机数字表法分为健康组10只,实验组Ⅰ、实验组Ⅱ、对照组各40只。实验组Ⅰ、实验组Ⅱ和对照组制成30%全身体表面积(TBSA)Ⅲ度烫伤模型。每隔1日晨8时,实验组Ⅰ小鼠1,25-(OH)2VitD31μg·kg-1+0.6 mL花生油灌胃;实验组Ⅱ1,25-(OH)2VitD34μg·kg-1+0.6 mL花生油灌胃;对照组0.6 mL花生油灌胃。分别于伤后1、3、7、14 d测空腹血糖(FBG)、空腹胰岛素(FIns)、血清肿瘤坏死因子(TNF)-α浓度和创面组织核因子(NF)-κB阳性率。结果 (1)实验组Ⅰ、实验组Ⅱ和对照组各时点胰岛素抵抗指数(HOMA-IR)、血清TNF-α水平和创面组织NF-κB表达阳性率均数均高于健康组水平。(2)实验组Ⅰ和实验组Ⅱ伤后同一时点HOMA-IR均低于对照组,且实验组Ⅱ低于实验组Ⅰ(P<0.05);同一组内不同时点伤后第3天HOMA-IR均数最高,以后逐渐降低(P<0.05)。(3)实验组Ⅰ和实验组Ⅱ伤后同一时点血清TNF-α水平、创面组织NF-κB表达阳性率均低于对照组,且实验组Ⅱ低于实验组Ⅰ(P<0.05);同一组内不同时点血清TNF-α水平、创面组织NF-κB表达阳性率均逐渐降低(P<0.05)。结论 1,25-(OH)2VitD3能够减轻严重烫伤小鼠应激状态下胰岛素抵抗作用和炎症反应。
【Abstract】 Objective To explore the effect of 1,25-dihydroxyvitamin D3[1,25-(OH)2VitD3] on insulin resistanceand inflammatory response in mice with severe burns under stress. Methods One hundred and thirty mice were randomlydivided into healthy group(n = 10), experimental group Ⅰ(n = 40), experimental group Ⅱ(n = 40) and control group(n =40). The model mice were scalded to 30% of total body surface area(TBSA) burnt(Ⅲ°) in experimental group Ⅰ, the experi-mental group Ⅱand control group. Mice in experimental groupⅠ were given by gavaging 1,25-(OH)2VitD3(1 μg·kg-1) with0.6 mL peanut oil at 8 am for every other day. At the same time and by the same way, mice in experimental group Ⅱ received1,25-(OH)2VitD3(4 μg·kg-1) with 0.6 mL peanut oil, and mice in control group received only 0.6 mL peanut oil. The serumlevels of fasting blood glucose(FBG), fasting insulin(FIns), TNF-α concentration, and the NF-κB positive rate in wound tis-sues were determined respectively on day1, day3, day7 and day14 after burn. Results(1) The values of insulin resistanceindex(HOMA-IR), serum TNF-α and the NF-κB positive rate in wound tissues were higher at different time points in exper-imental groupⅠ, experimental groupⅡand control group than those in healthy group.(2) The levels of HOMA-IR at thesame time points were significantly lower in experimental group Ⅰand experimental groupⅡ than those in control group, andthe value was significantly lower in experimental group Ⅱthan that of experimental group Ⅰ(P<0.05). The level of HOMA-IR was the highest at day 3 and then gradually decreased at the different time points in the same group(P<0.05).(3) The se-rum levels of TNF-α and NF-κB positive rate of wound tissues at the same time points were significantly lower in experimen-tal group Ⅰand experimental group Ⅱ than those in control group, and the value was significantly lower in experimentalgroup Ⅱthan that of experimental groupⅠ(P<0.05). The serum level of TNF-α and NF-κB positive rate of wound tissueswere gradually decreased at the different time points in same group(P<0.05). Conclusion 1,25-(OH)2VitD3can reducethe insulin resistance and inflammatory response in mice with severe burns under stress.
