宁夏枸杞总黄酮对人胃癌SGC-7901细胞增殖及细胞凋亡的影响

【摘要】 目的探讨宁夏枸杞总黄酮对人胃癌SGC-7901细胞增殖及细胞凋亡的影响。方法 MTT法检测宁夏枸杞黄酮(TFL)对人胃癌SGC-7901细胞增殖的抑制作用,Hochest染色观察TFL对人胃癌SGC-7901细胞形态的影响,流式细胞检测TFL对人胃癌SGC-7901细胞周期及细胞凋亡的影响。结果 TFL对人胃癌SGC-7901细胞具有抑制作用,48h的IC50为170μg/ml;TFL可引起SGC-7901细胞出现凋亡形态变化,并表现为剂量依赖;流式细胞检测证实TFL可以阻滞细胞周期及诱导细胞凋亡。结论宁夏枸杞总黄酮能够抑制人胃癌SGC-7901细胞增殖,阻滞细胞周期,诱导胃癌细胞凋亡,实验结果为进一步研究宁夏枸杞抗胃癌作用及其机制提供实验依据。

【Abstract】 Objective To discuss effects of the total flavonoids of Lycium barbarum L.(TFL) on the proliferation and apotosis of human gastric cancer SGC-7901 cells. Methods The inhibitory effects of TFL on proliferation of human gastric cancer cell line SGC-7901 was detected by MTT method; The influences of TFL on cell morphology of human gastric cancer SGC-7901 cells was observed by Hochest dyeing; The effects of TFL on cell cycle and cell apoptosis of human gastric cancer SGC-7901 cells was detected by Flow cytometry. Results TFL could inhibit the proliferation of human gastric cancer SGC-7901 cells,IC50 of TFL on SGC-7901 cells at 48 h was 170 μg/ml; TFL could cause the apoptotic morphologicai changes of SGC-7901 cells,and showed the dose dependent; Flow cytometry results showed that TFL can block the cell cycle and cell apoptosis. Conclusion TFL could inhibit the proliferation of human gastric cancer SGC-7901 cells,blocked the cell cycle and induced the cell apoptosis,this finding may provide an esperinental basis for further exploration of antitumor effect of TFL and its mechanism.
