大鼠血浆中芍药内酯苷、咖啡酸及绿原酸类浓度的UPLC-MS法同时测定及其药动学研究
Simultaneous determination and pharmacokinetics of albiflorin,caffeic acid and chlorogenic acid in rat plasma by UPLC-MS
【摘要】 目的建立UPLC-MS法同时测定大鼠静脉注射辛芍提取物后血浆中芍药内酯苷、咖啡酸、绿原酸、隐绿原酸、新绿原酸5种成分的分析方法,并计算5种成分在大鼠体内的药动学参数。方法色谱采用Waters BEH C18(2.1 mm×50mm,1.7μm)柱,流动相为0.1%甲酸乙腈-0.1%甲酸水梯度洗脱,质谱采用电喷雾电离源(ESI),扫描方式为选择性离子监测(SIR)。结果 5种成分在大鼠血浆中线性关系良好,提取回收率在95.1%~98.5%之间,日内、日间精密度、准确度和稳定性良好。各检测成分的曲线下面积[AUC(0~140 min)],表观分布容积(Vl),分布半衰期(t1/2α),消除半衰期(t1/2β)分别为53.71~867.55(mg/L·min),3.27~103.62 L/kg,15.15~24.08 min和23.79~35.96 min,结果表明辛芍提取物在体内吸收较为良好、分布较为广泛,且分布消除均较迅速。结论该方法可特异、快速、准确、灵敏地同时测定芍药内酯苷等5种成分在大鼠体内的经时血药浓度。
【Abstract】 Objective To establish a sensitive and reliable method for simultaneous determination of albiflorin,caffeic acid,chlorogenic acid,implicit chlorogenic acid and new chlorogenic acid in rat plasma using ultra performance liquid chromatography tandem mass sepctrometry( UPLC-MS) after the intravenous injection of the extract from Xinshao. Methods Five compounds were simultanously determined by UPLC with Waters ACQUITY UPLC BEH C18 column. The quantitative analysis of albiflorin,caffeic acid,chlorogenic acid,implicit chlorogenic acid,new chlorogenic acid and puerarin( internal standard) were carried out by UPLC with Waters ACQUITY UPLC BEH C18 column. The flow rate was 0. 35 ml /min. The mobile phase included 0. 1% formic acid in acetonitrile and 0. 1% formic acid in pure water. Electrospray ionization( ESI) and selective ion monitoring( SIR) were used as the source and scanning mode. Results Good linearity was achieved for the five compounds. The extract recoveries were 95. 1 ~102. 5%,the precisions,accuracy,matrix effects and stability of the analytes meet the requirements. The AUC( 0 ~140 min),Vl,t1 /2α,t1 /2β,were 53.71 ~867.55( mg/L* min),3.27 ~103.62 L/kg,15.15 ~24.08 min and 23.79 ~35.96 min. All the results indicate that the five compounds can be absorbed well and distributed widely. And their distribution and elimination are rapid. Conclusion This method is effective,rapid,accurate and sensitive for the simultaneous analysis of the pharmacokinetic parameters of these five compounds in rat.
