【摘要】 目的建立柱前衍生化高效液相色谱法分析不同生长年限金钗石斛多糖的单糖组成。方法将金钗石斛多糖样品用三氟乙酸(TFA)水解成单糖后,通过1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生,衍生物采用高效液相色谱梯度洗脱,研究金钗石斛多糖的单糖组成。结果不同生长年限金钗石斛多糖的单糖组成差异较大,一年生多糖由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、木糖、阿拉伯糖7种单糖组成,其摩尔比为68.33∶1.11∶1.00∶18.96∶3.44∶5.63∶7.26;二年生多糖由甘露糖、半乳糖醛酸、葡萄糖、半乳糖、木糖、阿拉伯糖6种单糖组成,其摩尔比为5.54∶1.07∶26.26∶1.00∶3.15∶5.75。结论该方法具有简便、结果准确等特点,可用于金钗石斛多糖中单糖组成的分析。该研究为金钗石斛的质量评价和开发利用提供了实验依据。更多还原

【Abstract】 Objective To develop a method based on precolumn derivation HPLC to analyze the monosaccharide compositions in polysaccharides of Dendrobium nobile Lind. from different growth years. Methods The polysaccharide were hydrolyzed into monosaccharides with TFA. The monosaccharide derivatives obtained with 1- Phenyl- 3- methyl- 5- pyrazolone( PMP) were sepa-rated by HPLC using a gradient elution process. Results The monosaccharide compositions in polysaccharides of Dendrobium nobile Lind. from different growth years were significantly differrent. 1- yeared polysaccharides was composed of mannose,rhamnose,galacturonic acid,glucose,galactose,xylose,and arabinose,with a molar ratio of 68. 33 ∶ 1. 11 ∶ 1. 00 ∶ 18. 96 ∶ 3. 44 ∶5. 63∶ 7. 26; 2- yeared polysaccharides was composed of mannose,galacturonic acid,glucose,galactose,xylose,and arabinose,with a molar ratio of 5. 54∶ 1. 07∶ 26. 26∶ 1. 00∶ 3. 15∶ 5. 75. Conclusion This method has accurate result and good reproducibility and can be used to determine the monosaccharides in polysaccharide of Dendrobium nobile Lind.
