辛夷中辛夷脂素与木兰脂素高效液相色谱含量测定方法的建立与优化

【摘要】 目的建立并优化辛夷中木兰脂素与辛夷脂素的含量测定方法。方法采用HPLC法同时测定辛夷中木兰脂素与辛夷脂素的含量,色谱柱为hypersil ODS2 C18柱(5μm,250×4.6 mm),流动相为甲醇-水(60∶40),流速为1.0 ml·min-1,检测波长为278 nm。结果木兰脂素在30~600μg·ml-1浓度范围内线性关系良好(r=0.999 7),平均加样回收率为98.72%;辛夷脂素在20~400μg·ml-1浓度范围内线性关系良好(r=0.999 5),平均加样回收率为98.64%。结论该方法准确、简便、快速,适用于辛夷中木兰脂素与辛夷脂素的含量分析,可为辛夷药材及其制剂的质量控制提供参考。

【Abstract】 Objective To establish and optimize the method for determination of magnolin and fargesin in Magnolia biondii Pamp. Methods HPLC method was used. Analyses were carried out on Hypersil ODS2 C18( 250 mm ×4.6 mm,5 m) column. The mobile phase was acetonitrile- water at the flow rate of 1 ml·min- 1. The wavelength of detector was set at 278 nm. Results The standard curve was linear with the range of 30 ~ 600 g·ml- 1for magnolin,and the average recovery rate was 98. 72%. The standard curve was linear with the range of 20 ~ 400 g·ml- 1for fargesin,and the average recovery rate was 98. 64%. Conclusion The method is simple and rapid,and suitable to determine the content of magnolin and fargesin in Magnolia biondii Pamp. The method can be used for the quality control of Magnolia biondii Pamp. and related preparatin.
