COX-2和ERCC1在胃癌中的表达及其临床意义

【摘要】 目的:探讨DNA切除修复交叉互补基因-1(ERCC1)和环氧化酶-2(cyclooxygenase-2,COX-2)在胃癌中的表达水平,并分析两者表达是否与淋巴结转移存在相关性。方法:采用免疫组织化学法检测239例胃癌组织中的ERCC1和COX-2表达水平,分析两者与临床病理参数间的相关性。结果:ERCC1、COX-2蛋白表达与淋巴结转移显著相关(P<0.05),而与肿瘤大小、部位、浸润深度、肿瘤分化等无关(P>0.05)。COX-2在Hp感染阳性胃癌组织中表达率较高,阳性表达率为86.5%。ERCC1与COX-2两组蛋白表达不存在相关性。结论:ERCC1、COX-2蛋白参与了胃癌发展,可作为判定胃癌生物学行为的临床指标,然而两者之间不存在协同作用。

【Abstract】 Objective To investigate DNA excision repair cross-complementing gene-1(ERCC1) and cyclooxygenase-2(COX-2) expression levels in gastric cancer(GC) and evaluate the correlation between two proteins. Methods Immunohistochemistry was performed to detect the expression of ERCC1 and COX-2 in 239 GC cases, and analyzed the association between ERCC1 and COX-2 expression levels and clinicopathologic parameters.Results ERCC1 and COX-2 expression levels were significantly correlated with lymph node metastasis(P <0.05), while not associated with tumor size, location, depth of invasion and tumor differentiation(P > 0.05). The positive expression rate of COX-2 in GC patients with HP infection was 86.5%, higher than those without HP infection. No correlation was found between two proteins. Conclusion Both ERCC1 and COX-2 were related with tumor progression, which may serve as valuable markers for determining biological behavior in GC. However, they did not show positive interaction.
