不同硒源对高碘致小鼠氧化性损伤的干预作用
Intervention Effects of Different Selenium Sources on Oxidative Damage in Mice Induced by High Iodine
【摘要】 目的:比较纳米硒和亚硒酸钠对高碘致小鼠氧化性损伤的干预作用。方法:48只雄性昆明小鼠被随机分为:适碘组(50μg/L KIO3);高碘组(3 000μg/L KIO3);亚硒酸钠干预组(3 000μg/L KIO3+0.193 mg/kg Se Na2SeO3);纳米硒干预组(3 000μg/L KIO3+0.193 mg/kg Se NANO-Se)。4周后处死,分离血清、取肝和肾脏组织。检测谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)等指标。结果:与适碘组比较,高碘组的肝中MDA含量下降,肝、肾、血清中GSH-PX活性均明显下降,肝、肾组织脏器系数均增加(P<0.05)。与高碘组比较,亚硒酸钠干预组的肾中MDA含量明显增加,肝、血清中GSH-PX活性明显增加,肝组织脏器系数下降;纳米硒干预组的肝、血清中MDA含量均明显下降,但肾中明显增加,肝、肾、血清中GSH-PX活性均明显增加,肾组织脏器系数下降(P<0.05)。与亚硒酸钠干预组比较,纳米硒干预组肝、血清中MDA含量均下降,肾中GSH-PX活性增加,肝组织脏器系数增加(P<0.05)。结论:本实验条件下,3 000μg/L的高碘摄入致小鼠氧化性损伤,应用0.193 mg/kg Se的亚硒酸钠和纳米硒干预后,对高碘致氧化性损伤均具有拮抗作用,纳米硒的干预效果明显高于亚硒酸钠。
【Abstract】 Objective: To compare the intervention effects of nano selenium and sodium selenite on oxidative damage in mice induced by high iodine. Methods: A total of 48 Kunming male mice were divided into 4 groups according to weight: control group( 50μg/L KIO3),high iodine group( 3 000 μg/L KIO3),high iodine( 3000 μg/L KIO3) plus sodium selenite( 0.193 mg/kg Se)group,high iodine( 3 000 μg /L KIO3) plus nano selenium( 0.193 mg/kg Se) group. After 4 weeks,related indexes including glutathione peroxidase( GSH-PX),superoxide dismutase( SOD) and malondialdehyde( MDA) were determined. Results: Compared with control group,in high iodine group,MDA content decreased significantly in liver,GSH-PX activity decreased significantly in serum,liver and kidney,tissue organ coefficient increased significantly in liver and kidney( P < 0. 05). Compared with high iodine group,in high iodine plus sodium selenite group,MDA content increased significantly in kidney,GSH-PX activity increased significantly in liver and serum,tissue organ coefficient decreased significantly in liver. In high iodine plus nano selenium group,MDA content decreased significantly in liver and serum and increased significantly in kidney,GSH-PX activity increased significantly in liver,kidney and serum,tissue organ coefficient decreased significantly in kidney( P < 0. 05). Compared with high iodine plus sodium selenite group,in high iodine plus nano selenium group,MDA content decreased significantly in liver and serum,GSH-PX activity increased significantly in kidney,tissue organ coefficient increased significantly in liver( P < 0. 05). Conclusion: High iodine intake may cause oxidative damage in mice. Selenium supplementation exert favorable effects on it. Intervention effect of nano selenium is significantly higher than that of sodium selenite.
