拮抗放线菌S-091菌株的发酵条件

【摘要】 为提高拮抗放线菌S-091菌株发酵液的抑菌活性,对其发酵培养的营养及环境条件进行研究。用番茄灰霉病菌作为抑菌活性测定的靶标菌,分别测定该菌株发酵培养基营养成分中不同种类的碳源、氮源、微量元素等物质以及发酵温度、培养基初始pH值、装液量、摇床转速等不同的发酵条件下菌株发酵液对靶标菌抑制作用的大小,以筛选出最适宜S-091菌株发酵培养的营养组成和环境条件。结果表明:在营养条件方面,可溶性淀粉和花生饼粉是菌株产抗生素的较优碳源和氮源营养;多种微量元素中KCl对菌株抗生素产生有明显的促进作用;在发酵环境条件方面,S-091菌株抗生素产生的较优培养温度为32℃,pH值为7,摇床转数为140r·min-1,适宜的装液量为250mL三角瓶装液100mL,最适宜的发酵时间为7d。

【Abstract】 For increasing the yield of antibiotics against plant pathogen from Actinomycete strains S-091, the fermentation condition of S-091 were studied. The activity of S-091 broth against the growth of Botrytis cinera was determined in relation to different nutriment component of substrate including some kinds of carbon sources,nitrogen sources,trace elements and in different condition of temperatures,shaking cultivation rpm, pH and volume of the broth. The result indicated that soluble starch and peanut cake were the best carbon source and nitrogen source respectively. Among 7 trace elements tested, KCl was the best one to promote S-091 producing antibiotics. The most optimum cultural conditions were initial pH7.5,capacity 100mL/250mL, temperature 32℃,140 r·min-1for 7d.
