免疫组化双染技术观察大鼠萎缩腮腺中肌上皮细胞的增殖变化
Proliferation of myoepithelial cells in rat atrophic parotid gland observed by double sequential immunohistochemical staining
【摘要】 目的:观察大鼠腮腺主导管结扎后腺体萎缩及肌上皮细胞(myoepithelial cell,MEC)的增殖变化规律。方法:对78只Wistar大鼠结扎右侧腮腺主导管,HE染色观察正常腮腺及主导管结扎后0(对照)、1、3、5、7、14、21、30、60、90、120、150、180 d腮腺的组织学变化(n=6),免疫组化双重染色方法定量分析MEC在腮腺萎缩不同时间点的增殖率及分布情况。结果:主导管结扎后,腮腺腺泡细胞逐渐凋亡消失,大量导管样结构形成,纤维组织增生并伴炎性细胞浸润。导管结扎后第3天,MEC快速增殖,5 d时增殖率达到峰值,7 d后MEC增殖率降低,30~150 d时MEC增殖率维持在一个较低水平,180 d时未观察到增殖的MEC,MEC总数显著减少。结论:结扎腮腺导管诱导腺体萎缩的过程中MEC早期增殖,随后增殖活力下降、停止。
【Abstract】 Objective: To observe the proliferation of myoepithelial cells(MECs) in atrophic parotid gland after main duct ligation. Methods: The right parotid duct of 78 Wistar rats was ligated,0(control),1,3,5,7,14,21,30,60,100,150 and 180 days after ligation the gland tissue was examined by HE staining and immunohistochemical double staining,the proliferation of MECs was analysed. Results: Apoptosis of acinar cells and formation of a large number of duct-like structures,fibrous tissue hyperplasia and inflammatory cells infiltration were observed after ligation. 1 ~ 5 days after duct ligation,the proliferation of MECs increased; 7 days after ligation,the proliferation rate decreased. 180 days ligation MEC stopt. Conclusion: MECs proliferate rapidly for a few days after ligation of parotid gland duct,with the atrophy of the gland the proliferation decreases,finally stops.
