目的:探讨利用慢病毒介导shRNA干扰mT OR信号途径,对口腔腺样囊性癌细胞生物学的影响。方法:shRNA干扰mT OR、雷帕霉素处理口腔癌ACCM细胞株,CCK-8法检测各组细胞的增殖情况,流式细胞仪检测各组细胞的凋亡率,Western blot法检测mT OR和Bcl-2蛋白的表达水平,real time PCR检测mT OR mRNA和Bcl-2 mRNA的表达水平。结果:细胞增殖情况,各不同刺激因素组与单纯ACCM细胞组比较增殖率降低(P<0.05)。不同刺激因素组细胞的凋亡率与单纯ACCM细胞组相比较,细胞凋亡率增高(P<0.05)。mT OR和Bcl-2的表达水平,雷帕霉素组和shRNA干扰mT OR组均低于单纯ACCM组(P<0.05)。结论:慢病毒介导的shRNA干扰信号途径,可影响口腔癌细胞的增殖,诱导细胞的凋亡,抑制了抗凋亡基因Bcl-2的表达,为肿瘤的靶向治疗提供了理论依据。
