三维能量多普勒超声检测宫颈癌的血流与miR-455表达的相关性研究

【摘要】 目的:探讨经阴道三维能量成像(3D-CPA)检测宫颈癌的血流与miR-455相对表达量的关系。方法:采用实时荧光定量PCR技术检测68例宫颈癌和49例慢性宫颈炎组织中miR-455的表达。用3D-CPA成像对宫颈血流进行检测并计算血管指数(VI),最后进行比较分析。结果:慢性宫颈炎组宫颈血流不丰富,VI为(0.439±0.181)条/cm3;宫颈癌组宫颈血流丰富、杂乱,VI为(0.940±0.447)条/cm3。宫颈癌组VI高于慢性宫颈炎组,P<0.01。宫颈癌组织内miR-455的表达量低于慢性宫颈炎组(1.17±0.59 vs 2.58±0.83,P<0.01)。宫颈癌组织内miR-455的表达水平与VI呈负相关(r=-0.295,P=0.014)。结论:miR-455低表达可能促进宫颈癌的血管生成,是一个潜在的抑癌基因。

【Abstract】 Objective:To investigate the correlation between three dimensional color power angiography(3D-CPA) and expression of miR-455 in cervical carcinoma.Methods:The expression of miR-455 in 68 cases with cervical cancer and 49 cases with chronic cervicitis(control group) was tested by real time PCR.All the cases were examined by transvaginal 3D-CPA preoperatively or before treatment.The vascular pattern and vascular index(VI) of the cervix were assessed.Results:The blood flow of the cervix was mainly abundant and disordered in cervical cancer group.While that was sparse in the control group.VI in cervical cancer group was significantly higher than that in chronic cervicitis group [which was(0.94 ± 0.447)/cm3and(0.439 ± 0.181)/cm3respectively,P < 0.01].The relative quantity of miR-455 in cervical cancer tissue was significantly lower than that in chronic cervicitis(1.17 ± 0.59 vs 2.58 ± 0.83,P < 0.01).Pearson correlation analysis showed that the expression of miR-455 was negatively correlated with VI in cervical carcinoma(r =-0.295,P = 0.014).Conclusion:Low expression of miR-455 may promotes the formation of microvascular in cervical cancer,and it may be a potential tumor suppressor gene.
