氟达拉滨联合STI571诱导K562细胞增殖的影响
Effect of K562 on Cell Proliferation Induced by Fludarabine and STI571
【摘要】 目的 针对氟达拉滨(Fludarabine)联合STI571诱导K562细胞增殖的影响进行探讨。方法 依据实验设计每12h依次检测浓度为5,10,20,30和40μmol/L的氟达拉滨以及与0.3μmol/L的STI571联合用药对K562细胞的抑制作用,总共4个时间段(12,24,36和48 h),并建立对照组。K562细胞增殖的变换通过MTT比色法进行判断,并比较氟达拉滨单用以及联合STI571使用的效果;药物的抑制率通过q=E(A+B)/[EA+EB(1-EA)]公式计算,q>1.15两药具有协同作用,q<0.85两药具有拮抗作用[注:E(A+B)代表两药联合使用的抑制率,EA,EB是两药单用的抑制率]。结果 随着氟达拉滨(Fludarabine)浓度逐渐升高,对K562细胞的抑制作用也逐步增加,细胞增殖抑制率也随之升高。各时间段数据经方差分析,差异有统计学意义(F值分别为16.56,18.57,15.9和19.56,P值均<0.01)。当不同浓度氟达拉滨(Fludarabine)与STI571联用36 h后,其q值分别为1.00,1.16和1.19,参照q值标准,说明两药联用具有协同作用。结论 氟达拉滨与STI571联用能明显增强其抑制K562细胞增殖能力,明显降低K562细胞的耐药性,对治疗效果有明显提高,指导临床治疗。
【Abstract】 Objective To investigate Fludarabine combined STI571 induced proliferation of K562 cells.Methods Designed each 12 h examines the density in turn for 5,19,20,30 and 40 μmol/L the chlorine farad shore as well as with 0.3 μmol/L the STI571 union medication based on the experiment,altogether 4 time section(12,24,36 and 48 h),and established the control group.By MTT colorimetric proliferation of K562 cells transform judgment,compared Fludarabine alone and combined effects of STI571 used;inhibition rate of drugs through q=E(A+B)/[EA+EB(1-EA)]formula computing,q>1.15,the two drugs had a synergistic effect,q<0.85,two drugs had antagonistic effects[Note:E(A+B)on behalf of the inhibition rate of the two drugs in combination,EA,EB were two drugs alone inhibition rate].Results Clofarabine while gradually increasing the concentration of K562 cells gradually increased cell proliferation inhibition rate increased.The F value respectively was 16.56,18.57,15.9 and 19.56(P<0.01).When different concentrations of clofarabine in combination with 36 h with STI571,its q values were 1.00,1.16 and 1.19,the standard reference value of q,indicating that the two drugs had a synergistic effect.Conclusion Fludarabine and STI571 combined with significantly enhanced its ability to inhibit proliferation of K562 cells was significantly lower resistance K562 cells,the treatment effect was significantly improved clinical treatment.
