【摘要】 目的 探讨前列腺干细胞抗原(prostate stem cell antigen,PSCA)、piwi样蛋白1(piwi-like 1,PIWIL1)和T-box转录因子2(T-box transcription factor 2,TBX2)的表达在肝细胞癌(HCC)中的临床意义。方法 收集2000~2003年HCC和相应癌旁正常组织所制备的石蜡切片各64例,应用免疫组织化学SP方法检测石蜡切片中PSCA,PIWIL1和TBX2蛋白的表达,分析三者的表达与HCC患者临床病理指标的关系。结果 在64例HCC的癌旁正常组织中,PSCA,PIWIL1和TBX2的表达率分别为17%(11/64),0%(0/64)和9%(6/64),而HCC中三者的表达率分别为73%(47/64),19%(12/64)和75%(48/64),PSCA,PIWIL1和TBX2的表达明显高于癌旁正常组织(X~2=40.859,13.241,56.505,P均<0.05);PSCA,PIWIL1和TBX2的表达均与患者性别、年龄无明显相关性(X~2=0.651~3.864,P值均>0.05),但是均与临床分期有关(X~2=12.923~39.511,P值均<0.05);PSCA的表达率随着HCC的病理分级而升高(X~2=18.047,P=0.000),而PIWIL1和TBX2的表达则与病理分级无明显相关性(X~2=3.356~5.688,P值均>0.05);除了PIWIL1以外(X~2=3.156,P=0.078),PSCA和TBX2的表达与淋巴结转移有相关性(X~2=6.409~8.466,P值均<0.05);PSCA和PIWIL1的免疫染色定位于肝细胞的胞浆中,而TBX2则定位于肝细胞的胞浆和胞核中。结论 PSCA,PIWIL1和TBX2在HCC中的表达研究为HCC发生发展分子机制提供了新的资料,PSCA和TBX2可能会成为治疗肝细胞癌的分子靶点。更多还原

【Abstract】 Objective To investigate the expression of PSCA,PIWIL1 and TBX2 in hepatocellular carcinoma(HCC).Methods The expression of PSCA,PIWIL1 and TBX2 Was detected with immunohsitoehemistry method in formalin-fixed,paraffinembedded HCC.Results The expression rates of PSCA,PIWIL1 and TBX2 were 17%(11/64),0%(0/64)and 9%(6/64)in 64 matched normal tissues respectively,but those were 73%(47/64),19%(12/64)and 75%(48/64)in 64 specimens of HCC.The expression rates of PSCA,PIWIL1 and TBX2 in HCC were statistica[iy higher than those in matched normal tissues,respectively(X~2=40.859,13.24 t,56.505;P<0.05).The expression of PSCA,PIWIL1 and TBX2 in HCC was all not correlated with gender and age,respectively(X~2=0.651~3.864,P>0.05),but it had association with stage,respectively(X~2=12.923~39.511,P<0.05).The expression rate of PSCA in HCC increased with pathological grade(X~2=18.047,P=0.000),but those of PIWlL1 and TBX2 did not(X~2=3.356~5.688,P>0.05).The expression of PSCA and TBX2 had correlation with lymph metastasis(X~2=6.409~8.466,P<0.05)except that of PIWIL1(X~2=3.156,P=0.078)in HCC.The immunoactivity of PSCA and PIWIL1 was located in cytoplasm of glandular cells and that of TBX2 was located in cytoplasm and cytoplasm/nuclei of glandular cells in liver tissues.Conculsion Reported the first analysis of PSCA,PIWIL1 and TBX2 expression in HCC and their implication for clinical features,PSCA and TBX2 might he candidate targets for the caner therapy.更多还原
