血卟啉单甲醚介导的声动力对U937细胞增殖影响

【摘要】 研究了聚焦超声激活血卟啉单甲醚对人髓系白血病细胞株U937细胞的增殖抑制作用.通过MTT法检测了频率为1.1MHz的聚焦超声激活血卟啉单甲醚作用于U937肿瘤细胞的存活率.MTT实验结果显示:声照辐射60s、强度为2~7 W/cm2的超声对U937细胞的抑制作用呈强度依赖性;单纯血卟啉单甲醚组与对照组相比无显著性差异(P>0.05);同等条件下,2 W/cm2超声结合10ng/L血卟啉单甲醚组对U937肿瘤细胞有明显的协同杀伤作用,与对照组相比有极显著性差异(P<0.01).这些结果表明,超声联合血卟啉单甲醚声动力能够特异性抑制U937细胞生长,血卟啉单甲醚有望成为一种新型声敏剂应用于白血病的声动力治疗.

【Abstract】 The present study aims to investigate the cytotoxic effect of focused ultrasound combining HMME on human leukemia U937 cells.Tumor cells suspended in serum-free RPM1640were exposed to ultrasound with 1W/cm2 and 1.1MHz for 60seconds in the presence and absence of HMME.The viability of cells was determined by MTT.MTT results showed that the treatment with ultrasound(1.1MHz,2~7 W/cm2,60s)alone significantly inhibited the cell proliferation of the U937cells in a intensity dependence,while the HMME(10~100ng/L) alone showed no statistically significant differences with the control groups(P>0.05);Under the same conditions,the synergistic effect of 2W/cm2 ultrasound combined with HMME showed significant cell killing compared with control(P< 0.01).These results demonstrated that ultrasound combined with HMME exerted specific inhibitory effect on the proliferation of U937 cells.HMME may be a promising sonosensitizing agent for the treatment of human leukemia.
