不同组培方式对花生子叶离体组培获得再生苗影响的研究
Study on Overcoming the Degeneration of Peanut by Tissue Culture Technology
【摘要】 花生栽培种在长期的种植过程中,往往由于多种因素的影响,造成种性退化,产量下降,品质变劣,抗病(逆)性降低,为了保持花生品种的固有特性,利用植物的组织培养技术,以花生种仁中的子叶作为外植体,采用MS培养基,附加不同种类和浓度的激素,进行愈伤组织的诱导培养,获得再生组培苗,再用不同的基质种植组培苗,搬到室外炼苗,约15d后移栽至大田。试验结果以混合NAA和2,4-D的培养基诱导率最高,达81%,基质以蛭石∶腐土=1∶1的幼苗成活率最高,达91%,幼苗室外移栽的最佳时期是3月份;大田定植期为4月上旬。
【Abstract】 Due to various factors,peanut cultivars got variety degeneration,yield and quality declined,and disease resistance(inverse)decreased in the long-term planted process.In order to maintain the inherent characteristics of peanut varieties,cultivation techniques were used by plant tissue,with peanut seeds cotyledons as explants.Induced culture of callus was carried out by applying MS culture base and hormones with different kinds and concentrations,in order to obtain regenerated plantlets.After planting in different substrates in vitro,the tissue culture seedlings were then moved outdoors and transplanted to the field 15days later.The test results showed the medium mixed with NAA and 2,4-D got high induction rate,up to 81%.The matrix:vermiculite rot soil=1∶1seedling survival rate was the highest,up to 91%.The best period of seedling field transplanting is March,and field planting period is in early April.
