HPLC法测定桑叶和沙棘叶中科罗索酸的含量
Determination of Corosolic Acid in Mulberry Leaves and Sea Buckthorn Leaves by HPLC
【摘要】 采用高效液相色谱法(HPLC)测定桑叶和沙棘叶中科罗索酸含量,为桑叶和沙棘叶进一步研究开发提供科学依据。色谱条件:Epic C18(4.6 mm×405 mm,5μm)色谱柱。在流动相甲醇∶0.2磷酸(V/V)为82∶18;流速为1 mL/min;检测波长为216 nm;柱温为30℃条件下测定桑叶和沙棘叶中科罗索酸含量。结果表明,科罗索酸在0.040 6~0.487 2 mg/mL质量浓度范围内,峰面积与浓度有着良好的线性关系;平均回收率为101.12%,相对标准偏差为1.074%(n=6)。HPLC法用于检测桑叶、沙棘叶中科罗索酸含量,简便易行、结果准确、重复性好。
【Abstract】 HPLC was used to determine the corosolic acid of mulberry leaves and sea buckthorn leaves in this experiments,it was provided a scientific basis for the mulberry leaves and sea buckthorn leaves in its further research and development. Chromatographic condition: Epic C18(4.6 mm×405 mm,5 μm)column was adopted. The mobile phase was methanol-0.2 phosphoric acid(82∶18).The detection wavelength was set at 216 nm. Flow rate was 1.0 mL/min. Column temperature was 30 ℃. The results showed that the calibration curve of corosolic acid was linear in the range of 0.040 6-0.487 2 mg/mL. The mean recovery of corosolic acid was 101.12%, RSD was 1.074%(n=6). The method was used to detect the corosolic acid of mulberry leaves and sea buckthorn leaves,simple, accurate and good repeatability.
