他克莫司后处理对缺血再灌注脊髓组织炎性反应的抑制作用
Inhibitive effect of tacrolimus postconditioning on inflammatory response provoked by reperfusion of ischemic spinal cord
【摘要】 目的探讨他克莫司后处理对大鼠脊髓缺血再灌注损伤后组织炎性反应的抑制作用。方法成年雄性SD大鼠90只,随机分为假手术(SO)组、缺血再灌注(IR)组和他克莫司后处理(TP)组。采用经股动脉置管球囊扩张制备脊髓缺血模型,缺血20分钟后抽出球囊中液体行再灌注。再灌注15分钟、1、6、24小时切取相应脊髓节段,采用邻联二茴香胺法检测髓过氧化物酶(MPO)活性,再灌注24小时应用干湿差重法测定伤段脊髓组织水含量,同时制备组织切片行原位末端标记法(TUNEL)染色观察神经细胞凋亡。结果 SO组大鼠脊髓组织MPO活性在7.20~8.03 U/mg之间,与之相比IR组大鼠再灌注各时间点MPO活性升高明显(P<0.01),TP组大鼠再灌注15分钟、1、6、24小时时MPO活性均显著低于IR组(P<0.01)。SO组大鼠脊髓组织水含量为(61.17±1.49)%,再灌注24小时时IR组大鼠高达(69.34±0.91)%,而TP组脊髓组织水含量为(65.09±0.61)%,显著低于IR组(P<0.01)。SO组大鼠脊髓神经细胞凋亡率为(5.44±1.31)%,再灌注24小时时IR组和TP组的凋亡率分别为(32.76±6.67)%和(20.40±5.43)%,TP组显著低于IR组(P<0.01)。结论他克莫司后处理能抑制缺血脊髓再灌注后的中性粒细胞聚集和组织炎性水肿,减轻神经细胞凋亡,从而发挥神经保护作用。
【Abstract】 Objective To explore the inhibitive effect of tacrolimus postconditioning on inflammatory response provoked by reperfusion injury of ischemic spinal cord in rats.Methods Ninety adult male SD rats were randomly divided into sham operation(SO)group,ischemia-reperfusion(IR)group and tacrolimus postconditioning(TP) group.The model of spinal cord ischemia was prepared by means of catheterization through femoral artery and balloon dilatation.Saline was removed out of balloon to reperfuse spinal cord 20 min after ischemia.The corresponding spinal cord segments were excised and measured for the activity of myeloperoxidase(MPO) by using dianisidine assay at 15 min,1,6,24 h after reperfusion respectively.Dry-wet weights method was employed to detected water content in injured spinal cord tissue,and TUNEL staining of tissue sections was applied to determine apoptosis of neural cells at 24 h after reperfusion.Results The MPO activities varied from 7.20 to 8.03 U /mg in spinal cord tissue of rats in SO group.The MPO activities in IR group were significantly higher than those In SO group at each time point after reperfusion(P <0.01),while the MPO activities in TP group were significantly lower than those in IR group at 15 min,1,6,and 24 h after reperfusion(P < 0.01).Water content in spinal cord tissue was(61.17±1.49) % in SO group,(69.34±0.91) % and(65.09±0.61)% in IR group and TP group respectively 24 h after reperfusion.Water content in TP group was significantly lower than that in IR group(P<0.01).The rate of neuronal apoptosis was(5.44±1.31) % in spinal cord of rats in SO group,and the rates were(32.76±6.67) % and(20.40 ± 5.43% in IR group and TP group respectively 24 h after reperfusion.The apoptosis rate was significantly lower in TP group than in IR group(P<0.01).Conclusion Tacrolinus postconditioning can suppress beutrophil accumulation,tissue inflammatory edema and apoptosis of neural cells to produce neuroprotective effect.
